Objrct:To observe the effect of Electroacupuncture on the regulation inflammatory response related factorsTNF-? and IL-4 in rats with cerebral ischemia reperfusion injury;To explore the mechanism of Electroacupuncture on improving the inflammatory response after cerebral ischemia in rats,so as to further deepen the understanding of the mechanism of Electroacupuncture in treatment of the disease and provide scientific theeoretical support basis for clinical treatment.Method:Chooosing male SD rats,according number table,randonly selected 24 for Sham group and the left remaining animals was established by modified Longa suture method becomed the cerebral ischemia reperfusion model.According to the criteria of the Zea-longa method,the initial judgment was successful or not.The successful models were randomly divided into Model group and Electroacupuntrue group,and the number of valid cases was 24.The Sham group was only isolated from the blood vessels without the insertion of the plug;Model group was blocked after the middle of the arterial occlusion of the arteries for 2h to achieve reperfusion,then do not do any treatment;Electroacupuncture group selected "Baihui" and "Dazhui" points for acupuncture,and connect the Korean electro-acupuncture instrument for 30min,while the waveform was continuous wave,the stimulus frequency was 2/15Hz.The neurological scores of each group were assessed by the Julio 's 18-point scale after ischemia-reperfusion for 24 hours.The morphological changes of cerebral ischemia and the percentage of cerebral infarct size were observed by TTC staining.The morphological changes of cerebral ischemia and the percentage of cerebral infarct size were observed by TTC staining,and brain tissue morphological changes were observed by hematoxylin-eosin(HE)staining.Immunohistochemical staining was used to detect the expression of IL-4 and TNF-? in defective cortical tissue.The expression of IL-4mRNA and TNF-? mRNA was detected by qRT-PCR.Result:(1)Neurological behavioral score:The neurological behavior score of rats with cerebral ischemia-reperfusion injury was significantly decreased,compared with the Sham group the difference was statistically significant(P<0.01);Compared with the Model group,electricacupuncture intervention can effectively improve the neurological score,the difference was statistically significant(P<0.05).(2)Changes of brain histomorphology and percentage of infarct size:Stained by TTC,the brain tissue of the Sham group showed dark red color,however the Model group showed white on the right side of the brain.The deletion area of Electricacupuncture group was lower than that of Model group.The percentage of cerebral infarction area in Model group and Electroacupuncture group was higher than that in Sham group(P<0.01);Compared with the Model group,the Electroacupuncture group's cerebral infarction area was significantly decreased(P<0.05).HE staining,the Model group sees a large number of shrunk cell aggregation,while the shrinking cells of the Electroacupuncture group were less than those in the Model group(3)Expression of IL-4 and TNF-a in cerebral cortex of ischemic rats:IL-4 protein expression:Compared with Sham group,the expression of histone and Electroacupuncture group were significantly increased,the difference was statistically significant(P<0.01);Compared with the Model group,the ecpression of Electricacupuncture group wai increased,the difference was statistically significant(P<0.05).TNF-a protein expression:Compared with Sham group,the expression of histone protein increased significantly,the difference was statistically significan(P<0.01),however,Electricacupuncture group was slightly increased,but the difference was not statistically significant(P>0.05);Compared with the Model group,the expression of Electroacupuncture group was significantly lower,the difference was statistically significant(P<0.05).L-4/TNF-a protein ratio:Compared with Sham group,the ratio of Model group depressed,but the difference was not statistically significant(P>0.05),while the ratio of Electroacupuncture group was significantly increased(P<0.01);Compared with the model group,the difference was statistically significant(P<0.01).(4)The expression of IL-4mRNA and TNF-? mRNA in the cerebral cortex of the defective side:IL-4mRNA expression:Compared with the Sham group,the Model group and the Electroacupuncture group were significantly increased,the difference was statistically significant(P<0.05 and P<0.01);Compared with the Model group,the expression of Electroacupuncture group was significantly increased,the difference was statistically significant(P<0.05).TNF-?mRNA expression:Compared with the Sham group,the expression of the Model group increased,the difference was statistically significant(P<0.01),but Electricacupuncture group has no statistically significant(P>0.05);Compared with the Model group,the Electricacupuncture group decreased,the difference was statistically significant(P<0.05).The ratio of IL-4/TNF-to mRNA:Compared with Sham group,the ratio of Model group was lowerr,but no statisticaliy significance(P>0.05),but there was significant difference between the two groups(P<0.01);Compared with the Model group,the difference was statistically significant(P<0.01).Conclusion:Electroacupuncture on IL-4 and TNF-a Inflammatory factors in rats with cerebral ischemia reperfusion injury was effected.enhancing the anti-inflammatory cytokines,down-regulate pro-inflammatory factors to made the ratio of IL-4/TNF-? enhanced.,and thereby reducing the inflammatory response in the ischemia-reperfusion zone,reducing the infarct area to improve neurological function.
