Effect Of Tongguan Decoction On The Expression Of TGF-1,Smad3 And CTGF Protein In SOI Model Rats

Objective:To investigate the effect of Tongguan decoction on the Expression of TGF-1,Smad3 and CTGF protein in Salpingitis Obstructive infertility model rats'fallopian tube tissue,Explore the mechanism of action Tongguan decoction and to provide basis for new drug development.Methods:The female SD rats SOI model was established by mixed bacteria with Staphylococcus aureus,Escherichia coli and Ureaplasma urealyticum.82 rats were randomly divided into 7 groups:the normal group,the sham operation group,the model group,the drug control group,the low dosage of Tongguan decoction(15.93g/kg·d^-1),the middle dosage of Tongguan decoction(31.86g/kg·d^-1)and the high dosage of Tongguan decoction(63.72g/kg·d^-1).After 30 days treatment,The rats were killed,Took the rats uterus andoviduct,observed the changes of the gross anatomy of the oviduct,After HE staining,Pathologic change of oviduct was measured by a microscope.the expression of TGF-1,Smad3 and CTGF protein in the oviduct was measured by Immunohistochemi-calmethod was used to detect.Result:1.The changes of Histomorphology and structure of oviduct in the model group were in accord with the change of oviduct inflammatory obstruction by naked eye and HE staining,Compared with the normal group and sham operation group,the difference was distinct,After treatment,the morphology and structure of oviduct were improved obviously,showed that the model wasbasically successful.2.the low dosage of Tongguan decoction,the middle dosage of Tongguan decoction and he high dosage of Tongguan decoctioncould improve the appearance of oviduct shape and color to a certain extent,gradually restore tubal ela sticity,lysis of fallopian oviduct adhesion with surrounding tissue,to promote the absorption of hydrosalpinx or empyemaby naked eye.the low dosage of Tongguan decoction,the middle dosage of Tongguan decoction and he high dosage of Tongguan decoction-could reduce the inflammatory cell infiltration,delay the proliferation of connective tissue,improve the morphology and structure of fallopian tube,increase the number of cilia and restore the functionby HE staining.3.the low dosage of Tongguan decoction,the middle dosage of Tongguan decoction and he high dosage of Tongguan decoctioncould reduce the expression of TGF-1,Smad3 and CTGF proteinin the oviduct tissue of model group?p<0.05,p<0.01?,improve the degree of fibrosis and restore the physiolo gical function of fallopian tube.Conclusion:1.The SOI model could be successfully prepared by inoculating the mixed bacteria in the vaginal cavity,Similar to human ascending infection,and was worth promoting.2.Tongguan decoction could reduce the inflammatory exudation of SOI model rats,reduce the swelling and congestion of the fallopian tube mucosa,promote the repair of the cells,improve the adhesion of the surrounding tissues,and accelerate the recovery of the fallopian tubes.3.The high expression of TGF-1,Smad3 and CTGF protein in the oviduct is involved in the pathogenesis of SOI,The possible mechanism of treatment of SOI by Tongguan decoctionis to inhibit the conduction of TGF-beta 1/Smads signal pathway,and thus to reduce the degree of fibrous scar in the fallopian tube.
