Objective:Through observing the influence of Huatanjieyu prescriptionson on 5-HT and NE in rats'brain, ACTH and CORT in rats'blood serum and morphological changes ofhippocampu, this thesis explores the mechanism of Huatanjieyu prescriptio nson onCUMS depression experimental rats with the purpose of providing objective t heoretial basis for clinical medication.Method:40 SPF level SD rats, half male and half female, were randomly divided into four groups:blank group, model group, positive control group and experimental group with 10 in each one. After seven days adaptive breeding, the blank group were treated freely to feed and drink, other groups were built as the CUMS depressed rats model for 21 days. Following the successful modeling, the blank group and the model group were treated with normal saline lavage, the positive control group was treated with fluoxetine, and the experimental group was continuously lavaged with Huatanjieyu prescriptionson q.d. Besides, every week the rats were weighed once and their ethology was observed. On the 43rd day, all rats were killed by drawing blood from femoral artery to test ACTH and CORT in ELISA detecting kit after centrifugation, namely, their brains were cut and then the brain tissues were immediately separated on the ice disc in order to test 5-HT and NE using the method of homogenate in ELISA detecting kit.Results:1. Three weeks after modeling, compared with the blank group, the model group, positive drug control group and experimental group were significant on the reduction of crossing scores and rearing scores, weight, sucrose intake and feed consumption (P<0.05); while there was no significant difference among the three groups (P>0.05).2. Six weeks after modeling, compared with the blank group, the model group was significant on reduction of crossing scores and rearing scores, weight, sucrose intake and feed consumption (P<0.05), while on the increase of ACTH and CORT in blood serum (P<0.05), and the reduction of 5-HT, NE in brain's tissue (P<0.05).3.Six weeks after modeling, compared with the blank group, the positive drug control group was significant on the increase or rise of crossing scores and rearing scores, weight, sucrose intake and feed consumption (P>0.05), and on the incr ease of ACTH, CORT in rats' blood serum (P<0.05), as wel as 5-HT, NE in rats' brain tissue (P<0.05)Compared with the model group, the positive drug control group was significant on the increase of crossing scores and rearing scores, weight, sucrose intak e and feed consumption (P<0.01), while on the reduction of ACTH, CORT in blood serum (P<0.01), and the increase of 5-HT, NE in brain tissue (P<0.01).4. Six weeks after modeling, compared with blank group, the experimental group showed no significant difference in crossing scores and rearing scores, weight, sucrose intake, feed consumption (P>0.05), as well as the ACTH, CORT in blood serum (P>0.05), and 5-HT, NE in brain tissue (P>0.05). Compared with the model group, the experimental group were significant on the increase of crossing scores and rearing scores, weight, sucrose intake, feed consumption (P<0.05), while on the reduction of ACTH?CORT in blood serum (P<0.05), and on the increase of 5-HT, NE in brain tissue (P<0.05).Conclusion:1.Through the increase of 5-HT and NE content in rats' brain, Huatanjieyu prescriptionson can play a therapeutic effect in the depressive order. Meanwhile, it also can cure the depressive order by the decrease of ACTH and CORT level in rats' blood serum to regulate the hyperactive HPA axis.2.Compared with the model group, the neurodegeneration of hippocampal tissue in Huatanjieyu prescriptionson experimental group is rarely observed, which indicates that Huatanjieyu has a protective effect on the hippocampal neuron of CUMS depression model rats.
