Experimental objective:To observe the San Zi Yang Qin decoction in asthmatic rats addicted to the expression level of eosinophil chemotactic factor (Eotaxin Eot-1, Eot-3), from the level of molecular biology to explore the mechanism of San Zi Yang Qin decoction to improve asthma symptoms.Experimental Methods:60 Wistar male rats were randomly divided into 6 groups, each group with 10 rats in each, compared with blank control group, asthma model group, the the San Zi Yang Qin decoction low dose group (SZ-L), middle dose group (SZ-M), high dose group(SZ-H), montelukast group (MLS). Except the blank control group, the other five groups of rats on day 1 and 8 days by intraperitoneal injection of ovalbumin (OVA) sensitized; from the 14th day, the five groups of continuous atomizing inhaled ovalbumin excitation 14 days (1 times/day,30min), in order to set up the model of asthma. In the blank control group, the sensitivity and stimulation of OVA were replaced by the same amount of normal saline. Rats in each treatment group were given oral administration of the first day of the excitation stage, and the time of filling the stomach was 1 hours before each stimulation. According to the different groups to fill the stomach During the course of the experiment, the attention was paid to observe the behavior changes of rats. The serum and lung tissue samples were collected from the rats after the last 24h excitation. The lung tissue was stained with HE, and the pathological changes of lung tissue were observed. The content of Eot-1 and Eot-3 in rat serum was detected by enzyme linked immunosorbent assay (ELISA), and the expression level of mRNA Eot-1 and mRNA Eot-3 in rat lung tissue was detected by RT-PCR technique.Experimental result:(1) pathology results:compared with normal control group, model group rat bronchioles are irregular in shape, see more inflammatory cell infiltration, thickening of the vascular wall, increasing the volume of lung appearance, surface a little swelling, irregular dark red hyperemic, asthma model was established successfully. Montelukast group and San Zi Yang Qin decoction each dose group rats lung tissue expression Pro:bronchioles morphological rules, and occasionally the infiltration of inflammatory cells and the lumen see less secretion, epithelial integrity, volume had no obvious change, surface hyperemia and bleeding obvious absorption traces, area is obviously less than the asthma group, bleeding quantity also decreased significantly. (2) The serum Eot-1 protein and Eot-3 protein content:?compared with the blank control group, model group rats serum Eot-1 content and Eot-3 protein increased significantly, the difference was statistically significant (P<0.05); ? compared with the model group, the content of each treatment group rats serum Eot-1 protein and Eot-3 protein is lower than that of model group, the difference was statistically significant (P<0.05); ?compared with the montelukast group, theSan Zi Yang Qin decoction high dose group rats serum Eot-1 protein, Eot-3 protein had no statistical significance (P>0.05);?San Zi Yang Qin decoction each dose groups, high dose the serum of rats in Eot-1 group and Eot-3 protein lower than low dose, the difference was statistically significant (P<0.05). ?San Zi Yang Qin decoction low dose group and middle dose group had no significant difference (P>0.05). (3) expression in lung tissue of rats with Eot-1mRNA and Eot-3mRNA: ?compared with the control group, the expression of Eot-1mRNA and Eot-3mRNA in serum of rats in model group were significantly increased, the difference was statistically significant (P<0.05); ?compared with the model group, the expression of each treatment group in the lung tissue of rats Eot-1mRNA, Eot-3mRNA lower than that of model group, the difference was statistically significant (P<0.05); ?compared with the montelukast group, the San Zi Yang Qin decoction high dose of Eot-1mRNA, Eot-3mRNA were not statistically significant (P>0.05); ?San Zi Yang Qin decoction each dose groups, the expression of the high dose group the serum levels of Eot-1mRNA, Eot-3mRNA lower than low dose, the difference was statistically significant (P<0.05). ?The San Zi Yang Qin decoction low dose group and middle dose group had no significant difference (P>0.05).Experimental conclusion:San Zi Yang Qin decoction can improve asthma rats lung tissue inflammatory infiltration and congestion and edema; San Zi Yang Qin decoction can reduce the expression of Eot-1 and Eot-3 in serum of rats. Eotaxin is involved in the inflammation of asthma, San Zi Yang Qin decoction could reduce the inflammatory cell infiltration in lung tissue of local through inhibition of the expression and release of asthmatic rats serum inflammatory factor Eot-1, Eot-3 and thus play inhibit airway inflammation, improve asthma symptoms.
