Polymorphisms Of Eotaxin-3 And The Association With Asthma

Objective To study the single nucleotide polymorphisms of +77 C/T and +2497 T/G in eotaxin-3 and their association with asthma, total IgE levels and eosinophil counts in adult population of the Han nationality from Hubei province of China.Methods Eosinophil counts with automatic blood cell counter, total IgE levels with chemiluminescent analyzer, eotaxin-3 +77 site and +2497 site amplification with polymerase chain reaction, the product of +77 site in eotaxin-3 was analyzed with restriction analysis and sequencing, the product of the +2497 site in eotaxin-3 with single strand conformation polymorphism, tetra-primer PCR amplification and sequencing.Results⑴Both T allele and G allele were detected at +2497 of the eotaxin-3; the frequency of the G allele in asthma group was 8.2%, as compared to 2.5% in control group; the frequency of eotaxin-3 +2497 G allele was significantly different between the two groups ( P<0.05). Both C and T allele were detected at +77; T allele in asthma group was 35.5%, and 36.5% in control group; the difference was not significant between the two groups (P>0.05). (2) For genotypes T/T and T/G at +2497 of the eotaxin-3 in asthma group, total levels of plasma IgE (lg IgE) were 2.73±0.31 and 2.91±0.32, the difference being significant between the two genotypes ( P<0.05). And C/C,C/T,T/T at +77 in asthma group, total levels of plasma IgE (lg IgE) were 2.74±0.32,2.75±0.32,2.85±0.29, the difference being not significant among the three genotypes (P>0.05). (3)The numbers of peripheral eosinophils were (0.36±0.28)G/L and (0.51±0.20)G/ L in genotypes T/T and T/ G at +2497 of eotaxin-3 in asthma group respectively, and the difference being significant between the two genotypes ( P<0.05). And the numbers of peripheral eosinophils were (0.44± 0.27)G/L,(0.37±0.27)G/L,(0.24±0.21)G/L in genotypes C/C,C/T,T/T at +77 in asthma group respectively, the difference being significant among the three genotypes (P<0.05).Conclusions Polymorphism of eotaxin-3 +2497 is associated with the susceptibility of asthma,peripheral eosinophil counts and total levels of plasma IgE in adult population from Hubei province, and polymorphism of +77 is associated with peripheral eosinophil counts.
