Objective:To discuss whether the P2X3 receptor mediates the anti-nociceptive effects of Moxibustion on inflammatory pain in C57BL/6J mices.Methods:1.All mices 20±2g were used in this experiments and were randomly di-vied into control group,model group(CFA group).Injected with complete Freun-d's adjuvant (20ul) to the hind paw for inducing inflammatory pain.Four days later,the model group were randomly divied into moxibustion group,CFA+A-3 17491 group,CFA+??-MeATP group,moxibustion+A317491 group,moxibustion+??-MeATP group;2.the moxibustion group were gived moxibustion 30min;the CFA+A-317491 group and CFA+??-MeATP group were injected A317491(P2X3 receptor antagonist) and ??-MeATP(P2X3 receptor agonist) into Zusanli acupuncture point (ST36) respectively;the moxibustion+A317491 group and moxibustion+??-MeATP group were applied moxibustion therapy 30min after injection;3.Determinated the pain threshold before CFA,4 days after model (before intervention),2 hours after the intervention;4.Detected the expression of P2X3mRNA that located in hypothalamus thr-ough Q-PCR;5.Statistic analysis was performed with the software 19.0 and figures were made by GraphPad Prism 5.0.Results:1.The analgesic effect of moxibustionCompared with the control group,the pain threshold of CFA group was great decreased after CFA injection (P<0.01);compared with the CFA group,there have statistical difference in the moxibustion group after moxibustion at 30min,60min,90min (P?0.05),obviously at 30min,then decreased gradually, there is no statistical difference at 120min,150min compared with before moxibustion.2.The influence of localized P2X3 receptor intervention on analgesic effect of moxibustion(1)The influence of the P2X3 receptor antagonist A317491Compared with the CFA group,the pain threshold of CFA+A317491group was increased at 30min,60min,90min (P<0.05);compared with the moxibustion group,both of them have anti-nociceptive effects,there are no difference (P>0.0 5);It is a great difference at 30mim after intervention (P<0.01) in the moxibu-stion+A317491 group,but lower than CFA+A317491 group and moxibustion gr-oup (P?0.05),and then decreased obviously at 60min,90min,120min,150min,co-mpared with after CFA and the CFA group,there is no difference (P>0.05)(2)The influence of the P2X3 receptor agonist ??-MeATPIt is no statistical difference of the thermal threshold in the CFA+??-Me-ATP group at all time points after CFA and compared with the CFA group (P> 0.05);There has statistical difference in the moxibustion+??-MeATP group after intervention, compared with the CFA+??-MeATP group and moxibustion group, especially at 30mim (P?0.05);But no difference with moxibustion group at 60 min,90min,120min,150min (P>0.05).3.The expression of P2X3 mRNA in the hypothalamusCompared with control group,the expression of P2X3 mRNA of CFA group is decreased (P?0.05); compared with CFA group,the expression of P2X3 mRNA of moxibustion group is increased (P<0.05).Conclusions:1.Moxibustion has a good analgesic effect in inflammatory pain with;2.The analgesic effect of moxibustion may require the participation of the localized P2X3 receptor;1)The analgesic effect of moxibustion was obviously reduced at the lack of P2X3 receptor;2)On the contrary, the anti-nociceptive effect of moxibustion was improved when ??-MeATP was injected to incitant the expression of P2X3 receptor.3.The P2X3 receptor in hypothalamus may be participate in moxibustion analgesic effect.
