The Discussion On Mechanism Of The Kidney-Warming Herbs Treating The Perimenopausal Syndrome

Perimenopause, which is a kind of physiological changes peculiarly belonging to women and a transition period marking the women’s reproductive function from strong to recession, refers the entire period from the menopause transition (45 years old or so) to 12 months after the menopause.Perimenopausal syndrome (PMS) means a series of physical symptoms, due to the menopausal women ovarian function decline and sex hormones to reduce, including the common sympotms such as hot flushes and sweating, irritability, palpitations and insomnia, menstrual disorders, etc.^.Western medicine is given priority to the sex hormone replacement therapy (HRT), however, the application is mianly subject to the side effects of HRT, by contrast, the traditional Chinese medicine is general accepted by the majority of patients owing to the better effect as good as the less side effects.On the basis of integrating ancient literature and modern clinical research, this paper argues that the kidney Yang deficiency,the kidney essence (tiangui) insufficiency, and the kidney Yang decline, cold and deficiency of Chong and Ren Channels, should be considered as the two main pathogenesis of perimenopausal syndrome.with the treatment requiring warming kidney Yang.The warming kidney yang not only promotes the tiangui metaplasia, but also provide continuous dynamics for tiangui playing physiological function by warming Chong and Ren Channels. As a result, this paper make a point according to the herbs of warming kidney Yang treating the disease, with taking tonic kidney essence as fundamental, taking warming kidney Yang and reconciling Chong and Ren Channels as kernel.To reveal the mechanism, this study,representing the Er-xian decoction, detect the Bcl-2, Bax protein expression and activity of Caspase 3 of follicular granulosa cells, by the method of enzyme-linked immunosorbent, for the sake of providing the experimental basis on the warming kidney Yang herbs treating the PMS.the results showed that: ①Er-xian decoction and its demolition parties all can inhibit the apoptosis of follicular granulosa cell;②the effect targets of Er-xian decoction and its demolition parties on inhibiting the follicular granulosa cell apoptosis are different: the Er-xian decoction group significantly degrades Bax protein expression and upgrade the Bcl-2 protein expression in follicular granulosa cells; the warmly invigorating kidney yang group and nourishing-yin purge-fire group significantly upgrade the Bcl-2 protein expression in follicular granulosa cells; the replenishing blood group significantly suppress Caspase-3 activity in the follicular granulosa cells, thereby preliminarily proving the mechanism of Er-xian decoction treating the PMS and providing theoretical basis of the warming kidney Yang herbs treating the PMS.This article will overall discusses the mechanism on the warming kidney Yang herbs treating the PMS from the traditional Chinese medicine theory, modern clinical medicine and pharmacology experiment.
