Plant growth regulators are pesticides which can regulate the growth of the plants though the external application. For the plant growth regulators could remarkably improve the growth as well as the yield and quality of the plants, they are being widely used in the agricultural production. However, if the anti season fruit was taken frequently, it may cause the metabolic disorders and various diseases and the surface residue has already influenced our country's food safety. Therefore, using simply,efficacy and property of pretreatment technology to detect the plant growth regulators residual in different matrix is meaningful to the food safety and the protection of the people's health.Objective:Our research was focus on the high-throughput methods those meet the domestic and international requirements in the sensitivity, accuracy, precision and the recovery though the advanced pretreatment methods and the UPLC-MS/MS or GC-MS/MS.The methods which we set up were used to analysis the plant growth regulators residual in fruits and vegetables that commercially available in Jilin province.Methods:Our research used the pretreatment technique and set up the methods of UPLC-MS/MS and GC-MS/MS which could qualitative rapidly and quantitative precisely in plant derived foods, respectively.Results:1. The UPLC-MS/MS method of 19 plant growth regulators which were Paclobutrazol,Gibberellin A3,(2,4-Dichlorphenoxy)acetic acid,thidiazuron,Forchlorfenuron,4-Chlorophenoxyacetic acid,3-indolyl-acetic acid,4-(3-indolyl)-butyric acid,6-Benzylaminopurine,N6-benzyl-9-(2-tetrahydropyranyl) adenine,6-Furfurylaminopurine, 2-Naphthoxy acetic acid, 2,3,5-Triiodobenzoic acid,trans-Zeatin,Daminozide,uniconazole,mepiquat chloride,Chlormequat chlorideV3-(3-indolyl)-Ipropionic acid in plant-based foods was established. The plant growth regulators residues were separated on a HSS T3(2.1 mm×100 mm,1.8?m) using acetonitrile and 5mmol ammonium acetate as the mobile phase. Good linarites were obtained in wide range and the correlation were greater than 0.997. The detection limits were between 0.02-5.0 ?g/kg. The rates of recovery were 72.3% to 115.8%and the relative standard deviations were 1.78%-5.32%.2. The GC-MS/MS method of 14 plant growth regulators which were4-Chlorophenoxyacetic acid,(2,4-Dichlorphenoxy)acetic acid, 1-Naphthyl acetic acid,2-Naphthaleneacetic acid,3-indolyl-acetic acid,4-(3-indolyl)-butyric acid,2,4-D-ethyl ester,2,4-D-1-butyl ester,1-Naphthyl acetamide,chlorflurenol-methy,Paclobutrazol, uniconazole, 6-Furfurylaminopurine, 6-Benzylaminopurine in plant-based foods was established. The plant growth regulators residues were separated on a DB-5MS column(30m×0.25 mm,0.25?m). Good linearity were obtained in wide range and the correlation were greater than 0.996. The detection limits were between 0.02-10.0?g/kg. The rates of recovery were 54.8%-131.6% and the relative standard deviations were 2.39%-6.38%.3. 223 commercially available samples include the watermelon, blueberry,waxberry, kiwifruit, grape, cherry, strawberry, bean sprout, tomato, cucumber etc. in Jilin province were detected using UPLC-MS/MS and GC-MS/MS methods as mentioned above during 2015-2016. The pollution data of the plant growth regulators in the plant derived foods of Jilin province were obtained. The results show that 14 plant growth regulators include Paclobutrazol, 6-Benzylaminopurine,trans-Zeatin, Chlormequat chloride, mepiquat chloride, Gibberellin A3,3-indolyl-acetic acid,(2,4-Dichlorphenoxy)acetic acid,4-(3-indolyl)-butyric acid,Forchlorfenuron,4-Chlorophenoxyacetic acid,2,4-D-ethyl ester,2,4-D-1-butyl ester,1-Naphthyl acetamide were detected, the detection rate was 56%.Conclusion:1. The research was set up detection methods of 19 plant growth regulators using UPLC-MS/MS. The method was rapidly and precisely, as well as the detection limit, recovery and the standard deviation were matched the standard of plant growth regulators residue requirement at home and abroad. And it could provide technical support to the safety supervise, the enforcement of the law and the respond ability topublic health emergencies.2. The research was set up detection methods of 14 plant growth regulators using GC-MS/MS. The method was fast, precisely, the detection limit, recovery and the standard deviation was matched the standard of plant growth regulators residue requirement at home and abroad. This method has the advantages of wide analysis range, faster analysis speed, less use of solvent, less pollution, low cost and easily to operate.. And it could be considered as a reference method to the safety supervise,the enforcement of the law and the respond ability to public health emergencies.3. There are certain food safety problems of plant growth regulators residue in the commercially available of the plant derived foods in our province, and our research provide the scientific basis to the further food risk assessment.
