Objective:Investigation of Paclobutrazol and other plant growth retardants (commonly known as Zhuanggenling), which are used in traditional Chinese Medicine by field investigation. Established the method for screening the residue.To know about the plant growth retardants residues in traditional Chinese Medicine.Methods:1)Investigation:through agricultural supplies sales shop, medicinal materials operating households farmers, government agricultural technology department, surveied plant growth retardants used type, usage history, time, amount, using the methods of investigation, medicine and chemical sampling, quantitative determination.Investigation of traditional chinese medicine and the areas are:Sichuan Province Taiping Town Jiangyou Pengzhou Zhang Ming Zhen aconite, Ao Ping Zhen chuanxiong, Rhizoma alismatis, Peng Shanxie Jia Zhen Mianyang Hua Yuan Zhen.2) An effective method was developed for screening Ophiopogonis Radix contamination by paclobutrazol and uniconazole using HPLC-MS/MS. The samples were pre-concentrated and purified using QuEChER sample preparation method. The separation was performed on a Thermo BDS HYPERSIL Ci8column with a gradient of acetonitrile-5mM/L ammonium acetate and0.5%acetic acid in pure water,detected by electrospray ionization mass spectrometry in positive mode with multiple reaction monitoring, quantified by external standard method.3) Determination of5Kinds of Plant Growth Regulator Residues in Traditional Chinese medicinal by HPLC-MS/MS:The samples were pre-concentrated with methanol. Useing a Waters Atlantis HILIC column as the separation column.Mobile phase gradient elution with a gradient of0.1%acetic acid5mM/L ammonium acetate solution:methanol: acetonitrile=13:15:72(V:V:V),detected by electrospray ionization mass spectrometry in positive mode with multiple reaction monitoring, quantified by external standard method.Results:The process of planting dwarf lilyturf root, Alisma, Achyranthes root, chuanxiong, radix rehmanniae, radix salviae miltiorrhizae, Radix Angelicae sinensis, pseudo-ginseng in their respective areas of different degrees are used of plant growth retardants.But plant growth retardant dosage, regulate the use of time rae not clear, the traditional chinese medicine residue did not take any attention. Some drugs containing plant growth retardants but not marked, may lead to excessive use of plant growth retardant agent. Simultaneous Determination of paclobutrazol and uniconazole in Ophiopogonis Radix by QuEChERS sample preparation method and HPLC-MS/MS:The optimized method offered a good linear correlation (R2>0.9957), excellent precision (RSD<7.88%) and acceptable recovery (from83.42to96.31%). Determination of5Kinds of Plant Growth Regulator Residues in Traditional Chinese medicinal by HPLC-MS/MS:Paclobutrazol, chlormequat, chloride, mepiquatchloride, choline chloride, daminozide showed a good linear relationship in the linear range respectively, correlation coefficient R2>0.996. Satisfactory recoveries, between79.26%and103.32%, at the three concentration levels assayed and excellent precision with RSD always lower thanl1%.Conclusion:This method of determination for paclobutrazol and uniconazole in Ophiopogonis Radix by QuEChERS sample preparation method and HPLC-MS/MS is proved to be simple and accurate.A effective HPLC-MS/MS method was developed for the separation and determination of5Kinds of Plant Growth Regulator Residues in Traditional Chinese medicinal,which can provide evidences to advanced researches in Pant Growth Regulator Residues.Plant growth retardant used in7kinds of Chinese herbal medicine planting process,...
