| Objective:Eczema is a kind of allergic dermatosis which is associated with a variety of internal and external factors.The study found that the main pathological mechanism is T helper cells imbalance and which caused cytokine secretion disorders.Interleukin-22(IL-22) and Aryl hydrocarbon Receptor(Ah R) as the important relevant cytokine secretion of Th22 cells play an important role in inflamamatory diseases and autoimmune diseases. The research will find the function of IL-22 and Ah R m RNA on the pathogenesis of eczema and the correlation IL-22 and Ah R m RNA with related EASI. The experiment preliminary discussion: 1. The expression levels differences of IL-22 and Ah R m RNA in peripheral blood of eczema patients and normals. 2. The expression difference of IL-22 and Ah R m RNA between the patients of different genders; 3. The expression levels differences of IL-22 and Ah R m RNA in peripheral blood of different type of eczema patients. 4. The expression levels differences of IL-22 and Ah R m RNA in peripheral blood of eczema patients after conventional treatment for two weeks and four weeks. 5. The correlation between IL-22 and Ah R m RNA, which discuss the interaction of the two factors on the pathogenesis of eczema. 6. The correlation between the expression levels of IL-22 and Ah R m RNA in peripheral blood of EASI.Methods:In the study, we chose 60 cases of eczema patients come from the Tianjin Union Medicine Centre as the experimental observation and 40 cases come from the Gerneral Hospital of Tianjin Medical University as normal control group. In the experiment, RT-PCR method was used to analyze the expression levels of IL-22 and Ah R m RNA in peripheral blood of 60 cases eczema patients and 40 cases normal control. According to the EASI criteria, we calculate the lesion and severity score of60 cases eczema patients. The statistical software SPSS17.0 was used to process experimental data, with P<0.05 or P<0.01 was statistically significant.Results:1. The expression levels of IL-22 m RNA and Ah R m RNA in peripheral blood of patients with eczema was obviously higher than those of the normal control group(IL-22⊿CT: 2.57±0.53 vs 5.91±0.78; Ah R⊿CT:2.87±0.50 vs 6.53±0.78,both P﹤0.01)2. The expression levels of IL-22 m RNA and Ah R m RNA in peripheral blood of the patients of different genders showed no otherness(both P >0.05)3. The expression level of IL-22 m RNA and Ah R m RNA of acute, subacute and chronic eczema group increased in turn(IL-22⊿CT : 2.79±0.47, 2.63±0.49,2.28±0.51, respectively, Ah R⊿CT : 3.05±0.51, 2.99±0.47, 2.58±0.38,respectively),all differences were statistically significant(P ﹤0.05),except,acute group and subacute group compared with no statistical significance(P>0.05).4. After 2-week treatment, the expression levels of IL-22 m RNA and Ah R m RNA in peripheral blood of eczema patients were lower than their first visits(IL-22⊿CT:3.77±0.58 vs 2.61±0.55, Ah R⊿CT: 4.46±0.81 vs 2.91±0.50). After 4-week treatment,the expression levels of IL-22 m RNA and Ah R m RNA in peripheral blood of the patients were much lower than 2 weeks ago(IL-22⊿CT: 5.05±0.79 vs 3.78±0.62,Ah R⊿CT: 5.66±0.95 vs 4.46±0.84), all differences were statistically significant(P﹤0.01)5. The expression levels between IL-22 m RNA and Ah R m RNA in peripheral blood of the patients were positively correlated and differences were statistically significant(r=0.433, P=0.001).IL-22 m RNA and Ah R m RNA in peripheral blood of the patients both showed positively correlations with EASI score(*r=-0.302, *P=0.019;**r=-0.117,**P=0.374).Conclusions:1. The expression levels of IL-22 m RNA and Ah R m RNA in peripheral blood of eczema patients were higher than healthy controls, and the expression levels of IL-22 m RNA were positive correlation with EASI, these data indicated that IL-22 and Ah R might participate in the occurrence and development of eczema.2. With treatment for two weeks and four weeks, the expression levels of IL-22 m RNA and Ah R m RNA were both decreased, which indicated that IL-22 and Ah R might participate in the pathogenesis of eczema. IL-22 and Ah R was the important relevant cytokine secretion of Th22 cells. So it was infered that Th22 cells could also play an important role in the occurrence and development of eczema. |
