Objectives 1 The retrospective analysis of lipid abnormality in patients with coronary heart disease(CHD) in Tangshan City. 2 The investigation of the association between circulating mi RNA-221 and high-density lipoprotein cholesterol.Methods 1 These patients with acute coronary syndrome(ACS)(n = 5 319; male: 3117; female: 2 202) and healthy subjects(n = 25 197; male: 11 114; female: 14 083)between January 2009 and December 2014 were enrolled from Tangshan Gongren Hospital. Retrospective analysis of lipid profile was performed. 2 Healthy subjects(n =174; male: 90; female: 84) between November 2014 and November 2015 were enrolled.Total plamsa mi RNAs was purfied according to Ambion mir Vana mi RNA Isolation Proccdure. The levels of mi R-221-3p and mi R-191-3p in plasma were examined by q RTPCR. The relative expression level of mi R-221-3p was normalized to mi R-191-3p and reported as 2-?Ct.Results 1 The retrospective analysis on lipid profile of ACS patients: 1) HDL-C levels in ACS patients, aged 20~55, 56~65 and 66~80 years, were significantly lower than those of healthy subjects for both male and female(P < 0.001). 2) Among 5 319 ACS patients,there are higher prevalence of low HDL-C(55.48% vs 29.69%, ?2 = 1 298, P < 0.01),high TG(36.79% vs 24.43%, ?2=343.4, P < 0.01) and LDL-C(23.90% vs 22.74%, ?2 =3.270, P = 0.07) than healthy subjects. 3) Binary logistic regression analysis showed that the odds ratio for CHD risk was 3.51(95% CI, 3.20-3.86) in those subjects with isolated low HDL-C phenotype. 2 The relationship between circulating mi R-221-3p and HDL-C levels: 1) Circulating levels of mi R-221-3p in female group were higher than those in male group(W = 5 277, P < 0.001). 2) Circulating levels of mi R-221-3p in healthy subjects with low HDL-C level were lower than those with normal levels of HDL-C(W =2 855, P = 0.006).Conclusions 1 These subjects with low HDL-C level, especially isolated low HDL-C phenotype, increased the risk of CHD in Tangshan City. 2 The detection of both mi R-221-3p and HDL-C level can be used as tools for CHD risk stratification management.Our results suggest mi RNAs located on chromosome X may be aossciated with HDL-C level.
