Objective : To investigate the relationship among the polymorphisms of Toll-like receptor 1(TLR1) H.pylori infection and susceptibility to gastric cancer in Qingdao,Shandong Province in China.Methods: The samples were collected from 225 patients with gastric carcinoma who were diagnosed ath first time and 225 normal personsin Qingdao municipal hospital during the January 2013 and December 2014. DNA was extracted by genomic DNA purification kit and the rs4833095 polymorphisms of TLR1 were genotyped by the gene sequencing. The correlation of polymorphisms of Toll-like receptor 1(TLR1) and H.pylori infection and susceptibility to gastric cancer were computed after adjusting for age, gender, helicobacter pylori infection with multiariable Logistic regression models.Results: The frequencies of GG?AG and AA genotype on TLR1 rs4833095 in gastric cancer patients were 33.8%(76/225), 49.8%(112/225) and 16.4%(37/225),respectively;those in control group were 43.6 %(98/225), 48.0 %(108/225) and 8.4 %(19/225),espectively, significant differences were found between the two groups(?2=8.64,P=0.013).It was found that the compared with the GG genotype, GA+AA genotype carriers obviously higher rise to gastric cancer(OR=1.42,95% CI: 0.96~2.13). Joint effect analysis showed that the GA+AA genotype and H.pylori infectors had higher rise to get gastric cancer(OR=2.23,95% CI:1.28~3.86)compared with GG genotype carriers and non- H.pylori infection group.Conclusion: The polymorphism of rs4833095 loci in TLR1 and H.pylori infection is closely related to the risk of gastric cancer.
