Objective: To observe the expression levels of Notch3,Lgr5 and NF-κB proteins in human colorectal adenocarcinoma,colorectal adenoma and normal colorectal mucosa specimens,explore the relationship between their expression and clinicopathological parameters of colorectal adenocarcinoma.Methods: Immunohistochemical SP method was used to detect the expression levels of Notch3, Lgr5 and NF-κB proteins in 54 cases of colorectal adenocarcinoma tissue, 30 cases of colorectal adenoma tissue and 12 cases of normal colorectal mucosa tissue. Combined with the clinicopathological data of patients with colorectal adenocarcinoma, the differences in expression levels of three proteins in different tissues were compared.Results: The positive expression rates of Notch3 in colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa tissue were 63.0%, 26.7%, 8.3% respectively. Compared with adenoma group and normal group, the colorectal adenocarcinoma group was significant difference(P<0.05),there was no significant difference between adenoma group and normal group(P>0.05); The positive expression rates of Lgr5 in colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa were 57.4%, 36.7%, 16.7% respectively. Compared with adenoma group and normal group, the colorectal adenocarcinoma group was significant difference(P<0.05),there was no significant difference between adenoma group and normal group(P>0.05); The positive expression rates of NF-κB in colorectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa were 63.0%, 30.0%, 25.0% respectively. Compared with adenoma group and normal group, the colorectal adenocarcinoma group was significant difference(P<0.05),there was no significant difference between adenoma group and normal group(P>0.05). The expression levels of Notch3、Lgr5 and NF-κB were not associated with gender, tumor size, differentiation degree in colorectal adenocarcinoma patients(P>0.05), but there was a correlation between their expression and the presence of lymph node metastasis and Dukes stages in colorectal adenocarcinoma patients(P<0.05),the expression with lymph node metastasis were higher than that without lymph node metastasis(P<0.05),the higher the Dukes stages level, the stronger the degree of those expression(P<0.05). In addition,the expression of Notch3 was also not associated with age in colorectal adenocarcinoma patients(P>0.05), but there was a significant correlation between Notch3 expression and the presence of the infiltration depth in colorectal adenocarcinoma patients(P<0.05),the stronger the tumor infiltration depth, the greater Notch3 expression in colorectal adenocarcinoma(P<0.05). The expression of Lgr5 was also not associated with age and the tumor infiltration depth in colorectal adenocarcinoma patients(P>0.05);The expression of NF-κB was also not associated with positions of the tumor in colorectal adenocarcinoma patients(P>0.05), but there was a significant correlation between NF-κB expression and the presence of the tumor infiltration depth in colorectal adenocarcinoma patients(P<0.05),the stronger the tumor infiltration depth, the greater NF-κB expression in colorectal adenocarcinoma(P<0.05).The Notch3 expression in colorectal adenocarcinoma was positively associated with the Lgr5(r=0.325,P<0.05),but there was no correlation between Notch3 and NF-κB(P>0.05). There was no linear correlation relationship between Lgr5 and NF-κB(P>0.05).Conclusion:1. The expression levels of Notch3、Lgr5 and NF-κB in normal colorectal mucosa specimens colorectal adenomas and colorectal adenocarcinoma present an gradually increasing trend,which indicated that their expression may be correlated with the occurrence of colorectal adenocarcinoma.2. The expressions of Notch3、Lgr5 and NF-κB in colorectal adenocarcinoma were found to stronger associated with the lymphatic metastasis and Dukes stages. The higher expressions of Notch3 and NF-κB in colorectal adenocarcinoma were also found to infiltration depth. Therefore,to some extent,the levels of Notch3、Lgr5 and NF-κB proteins in colorectal adenocarcinoma may indicate the malignant degree of colorectal adenocarcinoma and determine the prognosis of patients,the high expressions of three proteins may be good molecular biological indicators for lymph node metastasis and Dukes stages in colorectal adenocarcinoma.3. The expression of Notch3 in colorectal adenocarcinoma was positively associated with the Lgr5.
