Aims:To investigate the expression of NAD+-dependent 15-hydroxyprostaglandin dehydrogenase(15-PGDH) and Cycboxygenase-2(COX-2) in colo-rectal adenocarcinoma, colorectal adenoma and normal colorectal mucosa specimens and their possible implications.Methods: Immumohistochemical staining was used to detect the expression of 15-PGDH protein and COX-2 protein in colorectal adeno-carcinoma (n=40), colorectal adenoma(n=20) and normal colorectal mucosa (n=20).Results: The expression rate of 15-PGDH in colorectal adenocarcinoma(15%) and colorectal adenoma(20%) was significantly lower than in normal colorectal mucosa (100%) (p<0.01). The expression rate of 15-PGDH in colorectal adenocarcinoma(15%) and colorectal adenoma(20%) was no significant(p>0.05).The low expression of 15-PGDH in colorctal adenocarcinoma was found to be unrelated to the histological differentiation , Dukes stages and occurrence of lymphatic(p>0.05).The expression rate of COX-2 in colorectal adenocarcinoma(80%) and colorectal adenoma(70%)was significantly higher than in normal colorectal mucosa(25%) (p<0.01) and enhanced with the decreased histological differentiation, the increased Dukes stages and occurrence of lymphatic(p<0.05). The expression of 15-PGDH was negatively correlated with the expression of COX-2(r=-0.544, p=0.000).Conclusions: The expression of 15-PGDH is absent or decreased in colorectal adenocarcinoma. The loss or reduction of 15-PGDH expression may occur in the early development of colorctal adenocarcinoma.15-PGDH is an inhibitor of colon tumorig- enesis.The expression of COX-2 is increased in colorectal adenocarcinoma and promote the progression of colorectal adenocarcinoma.
