ObjectiveTo investigate the expression and effect of TGF-?1/Smad and KLF15 in different age groups' rats peritonealMethodsEighteen male SD rats were divided into three groups:(1)the juvenile group( 1months,n = 6);(2) the youth group(8 months,n = 6);(3) the elder group(18 months,n =6).At the end of the study, all the rats were weighed and measuring blood pressure and humanely killed, collecting the vein blood to test blood biochemical indexes: the changes of blood sugar?blood lipid?hepatic function and renal function.The parietal peritoneum tissues of rats were harvested and stained by hematoxylin-eosin and senescence-associated-?-galactosidase.The morphology changes of partial peritoneum were examined by light microscopy. Mesenteric expression of TGF-?1?Smad2?Smad3and KLF15 m RNA were evaluated by RT-PCR.The expression of TGF-?1?p-Smad2/3and KLF15 protein expression were evaluated by Western Blot.Results1. Groups of rats with the increase of age, the number of peritoneal mesothelium cell was decreased(P<0.05), peritoneal thickness and the number of peritoneal vessels were significantly increased(P<0.05);In the Senescence-associated-?-galactosidase staining, the number of senescent cells was increased in groups of rats(P<0.05), There was significant difference between juvenile group and youth group or elder group(P<0.05),2. Groups of rats with the increase of age,TGF-?1?Smad2?Smad3 m RNA levels and the expression of TGF-?1 ? p-Smad2/3 protein in peritoneum were significantly increased, meanwhile KLF15 was significantly decreased(P<0.05), There was difference between juvenile group and youth group or elder group(P<0.05),3. Correlation analysis found: there were negatively correlated to the expression ofKLF15 protein and TGF-?1 protein(R=0.97,P<0.05),and to the expression of KLF15 protein and the number of senescence cells in the senescence-associated-?-galactosidase staining(R=0.86,P<0.05).ConclusionsThe expression level of KLF15 m RNA and protein in rat peritoneal tissues was associated and decreased with age, while TGF-?1 and p- Smad2/3 expression level was increased with age. KLF15 could inhibit rat peritoneal natural aging, down-regulation KLF15 may be through up-regulation of TGF-?1?p-Smad2/3.
