Yi Qi Yang Yin Huo Xue Decoction's Effection On MiR-133a Expression Level Of Diabetic Rats

Objectives By using the realtime-quantitative PCR(qRT-PCR) technology, expression of mi R-133 a in myocardial tissues was detected to discuss Yi Qi Yang Yin Huo Xue decoction's effect on type 2 diabetes mellitus rats myocardial protection.Methods Within 60 male Wistar rats aged 12 weeks, 10 were selected randomly as the normal group, the rest 50 as type 2 diabetic group. After success in establishment of DM model by fat-enriched and high-energy diet, together with lower dose of streptozocin(STZ)-treated,rats in the type 2 diabetic group was randomly divided into 5 groups,namely model group, dan shen pills(DSP) group, Yi Qi Yang Yin Huo Xue decoction high, medium and low dose group, 10 in each group. Rats in Chinese medicine group and DSP group received drug intervention for 5 weeks; those in blank group and model group were given corresponding dose physiological saline. All rats were killed 5 weeks latter and blood glucose(FBG), body weight(BW), glycosylated serum protein(GSP), and blood lipid were measured. myocardial tissues from left ventricular anterior wall were isolated. One part was made into slices,followed HE staining, the myocardial cell changes of renal pathology were observed through microscope. Another part was used to detect expression of mi R-133 a in myocardial tissue through q RT- PCR technology. All date were analyzed by using One-way ANOVA.Results 1 Successful establishment on of diabetic rat model: rats in diabetic group suffered from polydipsia, polyphagia, polyuria, weight loss and other disorders. The rats' blood glucose levels were more than 16.7mmol/l, according to the standard of type 2diabetic rats. 2 During the experiment, rats in the diabetic group appeared symptoms of metabolic disorders, BW increased, movement reduced sharply, accompanied with poor spirit. Chinese medicine group and DSP group rats' overall situation improved better than those in model group, and rats in Chinese medicine medium group were more significant.3 FBG and GSP: After establishment of diabetic rats model, diabetic group rats' FBG and GSP increased, compared with the normal group(P<0.05). After drug intervention, FBG and GSP obviously reduce(P < 0.05) in the drug intervention group, compared with model group; compared with DSP group, FBG and GSP reduced in Chinese medicine medium and low dose group(P<0.05); compared between each Chinese medicine group,FBG and GSP of Chinese medium group was lower than the other two groups(P<0.05).4 Blood lipid: Compared with model group, TC?TG?LDL obviously reduced in the drug intervention group, HDL obviously increased(P<0.05); compared with DSP group,both Chinese medicine group had a stonger effect on reducing TC?TG?LDL(P<0.05),but there had no significant difference between DSP group and Chinese medicine group on HDL(P>0.05). Compared between each Chinese medicine group, TC?TG?LDL in the medium dose group were lower than other two groups(P<0.05), but there was no significant difference between groups of HDL level. 5 Observation on myocardial tissue pathological morphology: observed by light microscope after HE staining, normal myocardial tissue structure was clear, myocardial cell neat rows, a short columnar parallel to each other, oval nucleus, was located in the central; in model group myocardial cell rowed loose disorder, myocyte hypertrophy, degeneration, even visible wavy change,cardiac rupture, visible and irregular nucleus size, myocardial interstitial inflammatory cell hyperplasia; compared with model group, pathological morphology in each treatment group improved significantly, especially in the Chinese medicine medium dose group improving significantly. 6 mi R-133 a expression level of myocardial tissue: in the treatment group the expression of mi R-133 a had the varying degree increased(P<0.05)compared with model group. Compared with DSP group, mi R133 a level slightly higher(P<0.05) in the Chinese medicine group; The comparison had no significant difference between traditional Chinese medicine groups(P>0.05).Conclusions 1 The Yi Qi Yang Yin Huo Xue decoction can improve glucolipid metabolism of the type 2 diabetic rats, reduce the type 2 diabetic rats myocardial injury. 2Yi Qi Yang Yin Huo Xue decoction medium dose is stonger than that of high dose group and low dose group, suggests that we should pay attention to the relationship of concentration-response in the clinical practice, on the purpose of the most appropriate doses to achieve the optimal therapeutic effect. 3 The Yi Qi Yang Yin Huo Xue decoction can increase diabetic rats myocardial mi R-133 a expression level, inhibit cardiomyocyte hypertrophy, and is better than DSP, suggesting that Yi Qi Yang Yin Huo Xue decoction protection on myocardial injury of type 2 diabetes may be done through the regulation on gene level.
