Objectives Through observing the effect of Yi Qi Yang Yin Huo Xue prescription on detecting the biochemical index, expression level of mi RNA-146 a, and the coronary artery pathological morphology of type 2 diabetes mellitus rats, we explored the protect effection of Yi Qi Yang Yin Huo Xue prescription on diabetic rats coronary artery.Methods 12 weeks of age 60 SPF male Wistar rats were randomly extracting 10 as blank control group, while the others were set up the model of T2 DM by feeding high-fat diet combined with intraperitional injection of a small amount STZ many times. After the model set up randomly divided the models into model control group, Danshen treatment group, low/medium/high dose Yi Qi Yang Yin Huo Xue prescription treatment groups according to random number table, kept treating 5weeks. Observed fasting blood glucose(FBG), postprandial 2 hours blood glucose(2h BG) changes before and after intervention groups. After the experiment, measured blood serum C-reactive protein(CRP), nitric oxide(NO)and endothelin1(ET-1)content in abdominal aorta by ELISA; Detect expression level of mi R-146 a in left anterior descending coronary artery by q-PCR; Coring front wall of hearts of the coronary artery to make pathological specimens after put to death in rats,make specimens, and observe the change of vascular endothelial tissue.Results 1 Compared with blank group, rats’ general condition in model group were depression, polydipsia, polyphagia, urorrhagia, diarrhea, decreased activity, slow in response; Compared with model group, the rats’ condition above were improved significantly in groups of DSP group and all Chinese medicine groups; Compared with DSP group, middle dose group were similar, low and high dose groups were polyphagia and diarrhea.2 Compared with that in blank group, the levels of fasting blood glucose and 2h postprandial blood glucose were increased in model group(P<0.05); Compared with the model group, DSP group and all Chinese medicine groups could significantly reduce the blood glucose levels and lower the weights(P<0.05); Compared with DSP group, low dose group’s higher weights had no statistical significance(P>0.05), fasting and postpran-dial 2hours blood glucose reduced without statistical significance(P>0.05); Middle dose group’s lower weights had no statistical significance(P<0.05), fasting and postprandial 2 hours blood glucose were reduced significantly(P<0.05); High dose group’s weights redused significantly(P<0.05), fasting and postprandial 2 hours blood glucose were increased significantly(P<0.05).3 Compared with blank group, the levels of the serum CRP and ET-1 were increased, while the NO levels were decreased in the rats of the model group(P<0.05);Compared with model group, the serum CRP and ET-1 level were decreased, while the NO increased in DSP group and all Chinese medicine groups(P<0.05). Compared with DSP group, the serum CRP and ET-1 were increased(P<0.05), while the NO levels were decreased in the rats of the low dose group(P<0.05); The serum CRP and ET-1 were decreased(P<0.05), while the NO levels were increased in the rats of the middle dose group(P<0.05); The serum CRP and ET-1 were increased(P<0.05), while the NO levels were decreased with no statistical significance in the rats of the high dose group(P>0.05).4 Compared with blank group, the level of mi R-146 a was increased in coronary arterys(CA) of the rats of the model group(P<0.05); Compared with model group, the mi R-146 a level were decreased in DSP group and all Chinese medicine groups(P<0.05);Compared with DSP group, the mi R-146 a were increased in CA of the rats of the low dose group(P<0.05); While the mi R-146 a levels were increased with no statistical significance in the rats of the middle dose group(P>0.05); The mi R-146 a levels were increased in the rats of the high dose group(P<0.05).5 After HE staining, blank group cells display texture clear, shape was normal,distinctive nuance, CA cells had no hyperplasia, smooth, in the film smooth muscle cell(smooth muscle cell, SMC) had no hyperplasia, hemal wall with no thickening, tube cavity expansion without hyperemia, no inflammatory cell infiltration. Model group of rat aortic intimathickening of the limitations, endometrial local protuberance, membraneshallow of smooth muscle cells visible hyperplasia, arrangement disorder, elastic plate arrangement irregular wavy, rarely appear rupture separation. DSP group rats coronary artery pathological changes had eased. Dose group organization form change compared with model group significantly reduced as DSP group did. Compared with model group, low and high dose groups’ change were not obvious and much heavier than DSP group.Conclusions 1 Yi Qi YangYinHuoXue prescription can reduce the body weights, degree of FBG and 2h BG of T2 DM rats;2 Yi Qi Yang Yin Huo Xue prescription can effectively reduce the T2 DM rats serum endothelin-1 and C-reactive protein content, improve the contentof nitric oxide,reduce inflammation, effectively protect the coronary artery;3 Yi Qi Yang Yin Huo Xue prescription can inhibit inflammatory factor mi R-146 a expression levels in coronary artery of T2 DM rats, and gave evidence of the protection of Yi Qi Yang Yin Huo Xue prescription to T2 DM rats coronaryartery from the Angle of epigenetics which possibly provides a new targets for traditional Chinese medicine to improve the treatment of diabetic cardiovascular disease;4 Yi Qi Yang Yin Huo Xue prescription can effectively improve inflammatory reaction, and protect coronary artery of T2 DM rats..
