OBJECTIVE:To observe the expression level of miR-155and miR-146a in patients with coronary heart disease in peripheral blood mononuclear cells, and to explore the relationship between morphology and atherosclerotic plaque.METHORDS:148patients were divided into four groups, including42cases of AMI group,38cases in UAP group,33cases in SAP group,35cases in CPS group. Real-time quantitative PCR was used to detect the expression of microRNA-155and microRNA-146a in PBMCs.The positive rate of TLR4on monocytes of PBMCs were examined by flow cytometry. According to the number extent、types and plaque morphology characteristics of coronary plaque were classified.Based on Gensini score standards to quantitative assess the extent of coronary artery disease.of which112cases underwent imaging of64slice spiral CT coronary angiography in coronary atherosclerotic plaque, according to the CT value is divided into the control group,soft plaque, fibrous plaque, calcified plaque.RESULTS:1. The expression level of miR-155in AMI group and UAP group was significantly lower than that in CPS group (P<0.05); the expression level of miR-146a in patients in AMI group and UA group were significantly higher than CPS group (P<0.05).2. Coronary artery plaque was the main type II and soft plaque in AMI group and UAP group; the main typeⅠ、Ⅲ and fibrous plaque、calcified plaque in the SAP group;the control group in CPS group.3. There are differences which the expression level of miR-155. miR-146a and the positive rate of TLR4between different plaques.4. The expression level of miR-155were negatively associated with the positive rate of TLR4(r=-0.393, P=0.018),the expression level of miR-146a were positively associated with the positive rate of TLR4CONCLUSION:1.The expression of miR-155and miR-146a is associated with the severity of coronary stenotic lesions calculated and plaque stability,miR-155is a protective factor in the formation of coronary heart disease,and miR-146a is a risk factor.2.The expression level of miR-155、miR-146a and the positive rate of TLR4have differences in different plaques,miR-155plays a role of negative regulation of TLR4, and miR-146a is regulating role.
