?Objective?To investigate the expression of SOX11 and CD200, analyze its value for dignosis and prognosi in small B Cell Lymphomas.?Methods?1. Part ? : specimens of 5 groups, RH, MCL, MALTlymphoma/NMZL, FL and CLL/SLL(each group contained 30 cases) were collected for the study, which were surgically resected and diagnosed by pathologists in Fuzhou General Hospital of Nanjing Military Command from June 2006 to November 2015. IHC staining was used for the detection of SOX11 and CD200 expressions and the expression of SOX11 and CD200 was analyzed between different gruops.2. Part ?: total RNA were extracted from specimens of 30 cases of MCL and 6cases of RH. m RNA of SOX11 was detected by RT-PCR to analyze the correlation between m RNA and protein.3. Part ? : all MCL cases were collected complete clinical information and follow-up datas. Correlation was analyzed between the expression of two proteins and clinical pathlogical parameters and prognosis. Additionally, the relationship between the level of m RNA and prognosis was investigated.?Results?1. 27 of 30(90%) cases of MCL were positive for SOX11, and all cases of RH,MALTlymphoma/NMZL, FL and CLL/SLL were negative. Among all MCL, one case expressed SOX11 without expressing Cyclin D1; The sensitivity and specificity of SOX11 for diagnosis of MCL were 90% and 100%, respectively.CD200 expressed in 100% of RH, 50% of MCL, 50% of MALTlymphoma/NMZL, 76.7% of FL and 70.0% of CLL/SLL. But there was no significant difference in CD200 protein expression between 5 groups. In addition,compared with small B cell lymphomas group, CD200 expression in RH group was lower(P < 0.001).2. The mean of SOX11 m RNA in 26 available MCL specimens was 7.69; the standard deviation was 3.94; the minimum was-2.02; the maximum was 15.38.SOX11 m RNA level and the expression of the protein were related positively(p=0.015, ?s=0.470). In the negative SOX11 protein group, the mean of SOX11 m RNA was 0.24; the standard deviation was 1.99; the minimum was2.02; the maximum was 1.74. In the positive SOX11 protein group, the mean was8.67; the standard deviation was 2.95; the minimum is 4.47; the maximum was15.38. The m RNA Level of SOX11 in positive SOX11 protein group was significantly higher than negative SOX11 protein group(p < 0.001).3. The average overall survival time of 30 cases with MCL was 36.2 months, the median overall survival time was 31 months(1-96 months). The overall survival time of the positive SOX11 protein group was longer than the negative SOX11 protein group(33 months vs 6 months), with the difference being statistically significant(p = 0.028). The score of SOX11 protein for MCL had positive correlation with the overall survival time(p=0.003, ?s=0.717). The level of SOX11 m RNA had positive correlation the overall survival time(p=0.008,?=0.701), and the correlation was very strong. The overall survival time of the positive CD200 protein group had no statistical difference, compared with the negative group(p = 0.101).?Conclusions?1. The sensitivity and specificity of SOX11 for diagnosis of MCL were 90% and100%, respectively. Using immunohistochemical staining for SOX11 can be helpful for diagnosis of MCL, especially for diagnosis of cyclin D1-negative MCL. CD200 had no definite value in differential diagnosis of MCL,MALTlymphoma/NMZL, FL and CLL/SLL. In addition, compared with RH, its expression was downregulated.2. The protein expression of SOX11 had positive correlation with the level of SOX11 m RNA. The stronger SOX11 protein expression was, the higher m RNA level was.3. Both of the protein and the m RNA of SOX11 were associated with the prognosis of patients with MCL. The stronger protein expression and/or the higher m RNA level mean the prompt MCL patients prognosis was better. The expression of CD200 had no value for prognosis in patients with MCL.
