1 ObjectiveDetect the expressions of NM23-H2, Beclin-1, Bcl-2 in colorectal adenocarcinoma, and analyze the relationship between NM23-H2, Beclin-1, Bcl-2 and clinical pathology, meanwhile, the correlation of three proteins, the relationship between three proteins and patient prognosis. Discuss the role of three proteins in colorectal adenocarcinoma.2 MethodsWe collected one hundred and twenty-four cases of colorectal cancer tissues, forty-two cases of metastasizing lymph node tissues and twenty cases of adjacent tissues, twenty cases of normal lymph node tissues, and made tissues chip respectively. Immunehistochemical method(SP method) was used to detect the expressions of NM23-H2, Beclin-1, Bcl-2 in colorectal tissues. Relevant clinical information was collected and follow-up patients by the out-patient review, phone and internet.3 Results1) The positive rates of NM23-H2 were lower in colorectal adenocarcinoma tissues than in adjacent tissues, and the positive rates of Beclin-1, Bcl-2 in colorectal adenocarcinoma tissues were higher than in adjacent tissues, the differences were statistically significant(P<0.05). The different expressions of NM23-H2, Bcl-2 in metastatic and non-metastatic colorectal adenocarcinoma tissues were statistically significant(P<0.05), The different expressions of Beclin-1 in metastatic and non-metastatic colorectal adenocarcinoma tissues were not statistically significant(P>0.05), The different expressions of NM23-H2, Beclin-1, Bcl-2 in colorectal primary adenocarcinoma and lymph node metastatic cancer tissues were statistically significant(P<0.05), and in lymph node metastatic cancer tissues and normal lymph node tissues were statistically significant( P<0.05).2) In colorectal adenocarcinoma tissues, the expressions of NM23-H2, Beclin-1, Bcl-2 with tumor location, the age and gender of patients had no statistical differences(P > 0.05), All expression of NM23-H2, Beclin-1, Bcl-2 with TNM staging had statistical differences(P < 0.05). The expression of NM23-H2, Bcl-2 related to pathological grading(P < 0.05). The expressions of Beclin-1 were associated with infiltration degree(P < 0.05).3) By using the spearman rank correlation analysis, we analyzed the mutual relevance of the NM23-H2, Beclin-1, Bcl-2 and their correlation with the clinic pathological factors in colorectal adenocarcinoma tissues. The expressions of NM23-H2 and Bcl-2, pathological grading, TNM staging, lymph node metastasis showed a negative correlation(P<0.05), and had no correlation with Beclin-1(P > 0.05).The expressions of Beclin-1 with infiltration degree and TNM stage was positively correlated(P<0.05), and had no correlation with lymph node metastasis and the expression of Bcl-2(P>0.05). The expression of Bcl-2 positively correlated with pathologic degree, TNM staging, and lymph node metastasis(P<0.05).4) Single factor analysis showed that pathologic grading, infiltration degree, lymph node metastasis, TNM staging, NM23-H2, Beclin-1, Bcl-2 and colorectal adenocarcinoma patients correlated with overall survival and progression-free survival(P<0.05), Multiple factors analysis showed that NM23-H2 had no correlation with overall survival and progression-free survival,Beclin-1 and TNM staging show correlation with colorectal adenocarcinoma patients overall survival and progression-free survival(P < 0.05), the Bcl-2 protein was correlated with colorectal adenocarcinoma progression-free survival.4 Conclusion1) NM23-H2 may be anti-tumor role in colorectal adenocarcinoma; NM23-H2 negatively correlates with Bcl-2, the antitumor function of the NM23-H2 may be related to apoptosis.2) Beclin-1, Bcl-2 promote colorectal cancer malignant developing. But the correlation analysis shows that Beclin-1, Bcl-2 has no relevance, it prompts they have no synergic effect.3) Multi-factor analysis show postoperative overall survival, progression-free survival have no correlation with NM23-H2, it prompts NM23-H2 is not as an independent risk factor for the prognosis of patients. Beclin-1, TNM staging have correlation with overall survival and progression-free survival of patients, Bcl-2 only has correlation with progression-free survival, TNM staging, Beclin-1 can be used as the independent risk factors for prognosis of patients, Bcl-2 can be used as independent risk factors of patients with recurrence and transfer.
