Objective: The colorectal carcinoma is one of the most common malignant tumors in our country. It is severely detrimental to people's health. The occurrence and development of colorectal cancer is closely concerned with multiple oncogenes as well as anti-oncogenes. The anti-oncogene Syk and nm23-Hl closely affect the occurrence , development, invasion and metastasis of colorectal cancer, but many studies about the relationship of their expression with the development of colorectal cancer were limited on single indicator, the results are possibly controversial. The research was to analyze the expression of Syk and nm23-Hl in colorectal cancer tissues using immunohistochemical method (SP), in association with the clinicopathological parameters. Result of the expression of Syk and nm23-H1 was used to determine the correlation between Syk and nm23-Hl, and the relationship of expression of Syk and nm23-Hl in the occurrence, progression, invasion and metastasis of colorectal cancer. The aim of this research was to find a simple, ecnomic but effective method of molecular pathology for patients with colorectal cancer by immunohistochemistry method (SP) and to analyze their relationship,in order to supply basic theory for colorectal study. Methods: We randomly collected 60 colorectal cancer samples which were resected by surgery and confirmed by pathology in the First Hospital of Qin Huangdao from January, 2007 to October, 2007. In these 60 patients, 31 cases were males, 29 cases were females, mean age was 56.4 years old; 25 cases suffered from colon cancer, 35 cases rectal cancer; 50 cases belonged to adenocarcinoma, 10 mucous adenocarcinoma; 33 cases were lymph node metastasis; According to Duckes standard of clinical staging, 10 cases belonged to A, 26 to B, 24 to C. Furthermore, We made 30 normal colorectal tissues far from tumor 5cm as control. In our study, we observed the expression of Syk and nm23-Hl in colorectal cancer by immunohistochemistry method and analysed their relationship in colorectal carcinoma.Results1 Expression of Syk in colorectal cancerNormal colorectal tissues had high expression of Syk, 29 in 30 normal colorectal tissues. Compared to normal tissues,19 in 60 colorectal carcinoma did show positive expression of Syk,And there was significant difference between the two groups (χ2=33.95,p<0.05). Only 6 colorectal carcinoma tissues in 33 patients with lymph node metastasis had Syk expression. 13 colorectal carcinoma tissues in 27 patients without lymph node metastasis had the Syk expression. The Syk expression was negatively correlated with lymph node metastasis(χ2= 6.16, p < 0.05 ).Furthermore,the expression of Syk was correlated with the invasion deepth of tumor (χ2=7.05, p<0.05),but no correlation was observed with tumor size (χ2=0.22,p>0.05),tumor location (χ2=0.27,p>0.05) and age (χ2=0.43,p>0.05).2 Expression of nm23-Hl in colorectal cancerThe results demonstrated that the expression of nm23-Hl in colorectal carcinoma(53.3%) was lower than that in normal colorectal tissues (93.33%),the difference was very significant (χ2=14.40,P < 0.01).The expression of nm23-Hl had correlation with the deepth of tumor invasion, the expression of nm23-H1 in serosa tissues (25.00%) was lower than those mucosa layer (77.78%) and muscle layer(45.46%),the difference was very significant (χ2= 10.68,P < 0.05).The expression of nm23-Hl had correlation with lymph node metastasis,the expression of nm23-Hl in cases without lymph node metastasis (70.37%) was higher than those with lymph node metastasis( 39.4%),the difference was significant (χ2 =5.73,P <0.05). The expression of nm23-H1 was correlated with Dukes stage, the expression of nm23-Hl in Dukes C (33.3%) was lower than those Dukes A(80.0%) and Dukes B (61.54%).there were significant difference between Dukes A,B and C,(χ2=7.42, P <0.05)。3 The correlation between Syk and nm23-Hl expression in colorectal carcinoma.The positive expression of nm23-Hl had 18 cases in the colorectal tissues of expression of the Syk. In control, the negative expression of Syk had 27 cases in the colorectal tissues of negative expression of the nm23-Hl. There was significant difference between the expression of Syk and nm23-Hl in the colorectal cancer tissues (χ2=19.15,P<0.05). A positive correlation was observed between Syk and nm23-H1 in colorectal cancer(r=0.492).Conclusions:1 The normal colorectal tissue had the higher expression of Syk , but the lower expression of Syk in colorectal carcinoma. The expression of Syk had association with lymph node metestasis invasion deepth and Dukes stage, but no correlation with age, gender, tumor size,location of the tumor and differentiation degree.The lower expression of Syk had relationship with development, progression, invasion and metastasis of colorectal carcinoma.2 The normal intestinal tissue had the higher expression of nm23-H1, but the lower expression of nm23-H1 in colorectal carcinoma. The expression of nm23-H1 had connection with lymph node metestasis,invasion depth and Dukes stage,but no correlation with age,gender,tumor size,location of the tumor and differentiation degree.The lower expression of nm23-H1 had relation with development,progression, invasion and metastasis of colorectal carcinoma.3 The expression of Syk in colorectal carcimoma had the higher expression of nm23-H1. There was a significant result in statistics. This result suggested that low expression of colorectal carcinoma may be cooperated with Syk and nm23-H1 in the course carcinogenesis, development, invasion and metastasis of colorectal carcinoma.
