Objective:In order to explore the effect of electroacupuncture at Shuigou(GV26)on neuroneal autophagy and the possible mechanism of action after cerebral ischemia reperfusion injure in rats.Methods:Modified Longa method was employed to establish the experimental rat models with middle cerebral artery occlusion reperfusion(MCAO/R).The reperfusion was performed at 24 h and 72 h after middle cerebral artery occlusion(MCAO).Neurological deficit and cerebral infarction volume ratio was measured to assess the improvement effect,the infarction volume was measured by TTC staining,the morphological feature of neuron was observed by HE staining,the formation of autophagosomes was observed by transmission electron microscope,while the expression of LC3 B,p62 and the proportion of Bcl-2 positive cells were examined to observe the effect of electroacupuncture on autophagy.Results:1.Compared with sham operated group,neurological impairment,infarction volume and nerve cell injury were seen in model group and electroacupuncture group.Compared with model group,electroacupuncture group showed alleviated neurological impairment(p<0.05),reduced infarction volume(p<0.05)and relieved nerve cell injury.2.There were number of autophagosome in nerve cell of model group by transmission electron microscope.Compared with model group,electroacupuncture group rats resulted in decreased of the presence of autophagosome.3.Compared with sham operated group,model group rats resulted in markedly up-regulation of LC3B-II/LC3B-I ratio(p<0.01).While,compared with model group,electroacupuncture group rats resulted in decreased of LC3B-II/LC3B-I ratio at 24 h and 72 h after reperfusion(p>0.05).4.Compared with sham operated group,model group rats resulted in markedly down-regulation of p62 at 24 h after reperfusion(p < 0.05).While,compared with model group,electroacupuncture group rats resulted in significant increased of p62 at 24 h after reperfusion(p<0.05).5.Compared with model group,electroacupuncture group rats resulted in markedly increased of the number of Bcl-2 positive cells at 24 h after reperfusion(p<0.01).Conclusion:1.Electroacupuncture at Shuigou could promote the recovery of neurological function and reduce the infarct volume after cerebral ischemia reperfusion injure.2.After cerebral ischemia reperfusion injury,the autopgagy could be induced in rats.And the result in this thesis demonstrated that the treatment of electroacupuncture at Shuigou(GV26)could prevent the autopgagy.3.The treatment of electroacupuncture at Shuigou(GV26)could protect nerve cell from autophagy may through up-regulation of the expression of p62 and Bcl-2,so that the inadequate of autophagy was alleviated.
