Purpose:Explore the influence of the electric needle to aim directly at the synaptic morphous of neurons after ischemic brain damage.Method:Select 27 male SD rats to randomly divide into 3 groups:the normal group,the model group,the electric needle group.Among,the normal group wasn't adopt any intervention study,the model group and the electric needle group adopt middle cerebral artery occlusion(MCAO) to make the model of focal cerebral ischemia.The electric needle group carried out electro-acupuncture treatment the next day after the model of focal cerebral ischemia was made successfully:We chose Baihui Point and Shuigou Point,and made use of the electro-acupuncture apparatus of G6805-2A type,continuous wave(CW),frequency of 20 Hz,intensity was to make the head shake lightly,30 minutes every time,once a day,took a rest in the middle,the course of treatment was two weeks.After treatment,according to the method of drawing materials from the normal position,drew the materials from hippocampal CA1 section which is the most impressionable to ischemia in the blood-supply district of middle cerebral artery.Through the procedure as follow:antero-fix→poaching→postfix→poaching→dewatering→soak→investing,polymerizing→sectioning,dyeing and so on. Observed and shot pictures under the electron microscope of Hitachi Hu-12Atype,drew assistance from the image analysis system of Motic Images Advanced 3.0 to survey the length of arc and chord of synaptic interface of GrayItype,calculate curvature of synaptic interface(length of arc/length of chord),and added up the quantity of GrayItype synapse every group.to compare the variance in the size of synaptic curvature of hippocampal neurons and in the quantity of GrayItype synapse every group.Result:(1) the model group(1.07±0.03) to compare with the normal group(1.16±0.03)in synaptic curvature had a very significant variance(P<0.01);(2) the electric needle group (1.12±0.05) to compare with the normal group(1.16±0.03)in synaptic curvature varied significantly(P<0.05);(3) the electric needle group(1.12±0.05) to compare with the model group(1.07±0.03) varied significantly(P<0.05);(4) The quantity of GrayItype synapse of three groups compared with each other had no significant variance.Conclusion:The numerus of the synaptic curvature of the model diminished significantly; after the treatment of electro-acupuncture,the synaptic curvature of the electric needle group increased,to hint that electro-acupuncture therapy can make synaptic morphology obtain a certain degree of repair,thereby to prevent hippocampal synapse cataplasia,promote synapse reconstruction,and improve the function of brain.But the bending type of synaptic interface had no significant variance in this experiment.
