Objective: This study investigated the RACK1,VEGF,SOX2 and p40 protein expression in non-small cell lung cancer tissues and to analyze their value in early diagnosis and clinical significance.Methods: In this study,87 cases of surgically removal primary non-small cell lung cancer tissue and 50 cases of normal lung tissue 95 cases of bronchoscopy biopsy tissue and 43 pleural effusion cytology specimens,obtained from the First Affiliated Hospital of Shan Xi Medical University between 2006 and 2012,To verify the morphologic diagnosis,the routine H&E-stained sections were reviewed.The expression of RACK1,VEGF,SOX2 and p40 protein in non-small cell lung cancer tissues were investigated by immunohistochemistry En Vision method.The relationship with the clinicopathological parameters were also analyzed.And compare the diagnosis value of the above four indicators with TTF-1 and p63 in non-small cell lung cancer.Results:(1)The positive rates of RACK1 and VEGF in non-small cell lung cancer tissues were significantly different with that in non-cancerous adjacent tissues(p<0.05),the positive expression rates of RACK1 and VEGF in lung adenocarcinoma were significantly higher than squamous cell carcinoma(p<0.05).The sensitivity and specificity of RACK1 protein in the diagnosis of lung adenocarcinoma were higher than VEGF.Its sensitivity was higher than TTF-1,however,its specificity was below TTF-1 in the diagnosis of lung adenocarcinoma.(2)The expression of RACK1 and VEGF were correlated with the clinical TNM stages and lymph metastasis of lung adenocarcinoma(p<0.05).The expression ofVEGF was correlated with the degree of differentiation in lung adenocarcinoma(p<0.05).Spearman rank correlation results indicated that the level of RACK1 expression correlated positively with VEGF protein expression(r=0.339,p<0.05).Survival analysis showed that survival time of the lung adenocarcinoma patients was significantly reduced in cases of patients with positive expression of RACK1 and VEGF(p<0.05).(3)The positive rates of SOX2 and p40 in non-small cell lung cancer tissues(62.6%vs 54.9%)were significantly different with that in non-cancerous adjacent tissues(p<0.05).The positive expression rates of SOX2 and p40 in squamous cell carcinoma were significantly higher than lung adenocarcinoma(p<0.05).The sensitivity and specificity of p40 protein were higher than SOX2 and p63 in diagnosis of squamous cell carcinoma.(4)The expression of SOX2 and p40 were correlated with the clinical TNM stages and lymph metastasis of lung squamous cell carcinoma(p<0.05).The expression of p40 was correlated with the degree of differentiation in lung squamous cell carcinoma(p<0.05).Spearman rank correlation results indicated that the SOX2 and p40 protein expression in squamous cell carcinoma tissues with no correlation(p>0.05).Survival analysis showed that survival time of the squamous cell carcinoma patients was significantly reduced in cases of patients with positive expression of SOX2 and p40(p<0.05).Conclusion:(1)RACK1 can be used as complements diagnostic markers with TTF-1protein in diagnosis of adenocarcinoma.RACK1 and VEGF protein may promote each other in the process of lung adenocarcinoma.The positive expression of RACK1 and VEGF may indicate poor prognosis in lung adenocarcinoma patients.(2)p40 is a diagnostic marker which is more sensitive and specific than SOX2 and p63 in squamous cell carcinoma.SOX2 and p40 may play an important role in the occurrence and progression of lung squamous cell carcinoma.The positive expression of SOX2 and p40 may indicate poor prognosis in lung squamous cell carcinoma patients.
