As a commonly used clinical Chinese medicine,Rehmannia glutinosa Libosch was first recorded in the Shen Nong’s Herbal Classic and was listed as one of the top grades.With regards to the evaluation ofquality of Rehmannia glutino,Li Shizhen once said,“Today’s people only regard the Rehmannia glutinos of Huaiqing as the best,meanwhile it differs with the rise and falls of ”.Huaiqing mainly refers to an area around Jiaozuo,Henan Province,including Wenxian,Wuzhi,Qinyang,Bo’ai,among others.Up to now,Rehmannia glutinos of Henan’s Huaiqing is still seen as the most genuine medicinal herb.Except Henan,Rehmannia glutinos is also cultivated in Shandong,Shanxi and other places.Taking the Rehmannia glu tinos produced in Henan,Shanxi and Shandong as an research object,this paper will conduct a chemical analysis,biological evaluation and near-infrared analysis for Rehmannia glutinos from different medical herb markets and places,discuss the relationship between commodity specification and its chemical composition and biological activity of Rehmannia glutinos and furthermore develop new product specifications to provide a scientific basis for quality control of Rehmannia glutinos.First of all,based on the standard in 76 kinds of medical products specifications published in 1984,this paper ranked Rehmannia glutinos according to its weight,surface color,section color and section texture,and as a result,the collected samples were divided into five ratings: there are 5 levels according to weight;2 levels according to surface colors(brown-black and brown-grey);4 levels according to section colors(brown-black section,claybank section,section with inner brown and outer yellow);3 levels according to section textures(general texture,cuticle texture,oily textur e)and we got 180 samples in the end.Taking iridoid glycosides(Catalpol,rehmaionoside A,rehmaionoside D,Motherwort glycosides),verbascoside,monosaccharides(glucose,fructose),oligosaccharides(sucrose,raffinose,water Su sugar),polysaccharides and amino acid content as an index,this paper conducted a test of chemical composition content for the 180 samples with the use of HPLC.At the same time,it detected water content,total ash,acid insoluble ash,water-soluble extract and analyzed experimental results by using variance analysis,cluster analysis and principal component analysis,etc.Analysis of variance showed: there is no obvious correlation between weight,surface color,section texture and its chemical composition of Rehmannia glutinos,while a certain regularity exists between section color and chemical composition that with the gradual deepening of section color,the content of iridoid ingredients glycosides and oligosaccharides decrease,monosaccharides and polysaccharides components gradually increase;principal component analysis and cluster analysis showed that: with the gradual deepening of section color,the comprehensive evaluation of Rehmannia glutinos quality lowers.In this paper,anti-oxidation and hypoglycemic activity of Rehmanniag lutinowas evaluated with the use of vitro model.We found: the antioxidant IC50 value of sample of of Rehmannia glutino was 1.96 ~ 3.43 g/L,less than the concentration of the positive control VE ethanol solution of3.67 g/L,which illustrated that of Rehmannia glutino had a strong anti-oxidation;sort order of grades and section colors showed that there is no significant correlation between antioxygenation and its weight,surface color and section texture of Rehmannia glutino,but there is some correlation with the section color and black section has the strongest antioxidant.In addition,it showed that of Rehmannia glutino only had a weak supportin grole in blood sugar decreasing effect.With the use of near infrared diffuse reflectance,this paper conducted aqualitative analysis on 180 rehmannia samples.The results showed that Rehmannia glutinos of the 1st level can only separate from other four leves in class model,the other four leves are in a mixed state.Which showed that it is impossible that using class analyzed samples;In the model of section color,in the first,it is to done to recognize and analyze four section colors,finding that section with inner brown and outer yellow and claybank section or tan section are in crossed state,the model is not good.When merges section color(outer yellow brown classified as claybank),it is founded that only two methods can separate these samples and the model is not stable and relibale.When merges section color(outer yellow brown classified as tan),it is founded that most methods can separate thes e samples and the model is stable,reliable,and samples can be better separated.
