Objective This study adopts the method of middle cerebral artery occlusion establishing rats model with cognitive impairment.Electric acupuncture at Shenting and Baihui,assessment of animal cognitive behavioral and learning and memory ability,using molecular biology techniques to detect autophagy related proteins and gene expression.To explore the possible mechanism of electric acupuncture at Shenting and Baihui to improve the effect of cognitive impairment and the relevance of autophagy regulation system.Methods 48 Sprague-Dawley male rats were randomly divided into 3 group:electric acupuncture group and ischemia control group,each of them 18 and sham operation control group12.By using the method of middle cerebral artery occlusion establishing focal cerebral ischemia reperfusion rats model of cognitive impairment.after 2h of operation,we applied EA at Shenting and Baihui,30 min once daily,a total of 10 days;Fetching model group and control group was on the same conditions,but no any treatments.Using Longa to assessment neural function defect in rats,Morris water maze assessment model rat spatial learning and memory ability;Western Blot in the validation of the left hippocampus of rats Beclin1,LC3I/II protein expression and rt-pcr technique to detect Beclin1,LC3I/II,Bcl2 gene expression levels.Results 1.The Longa score show that after surgery the sham operation group showed no neurologic deficits symptoms;Before the treatment,there are varying degrees of nerve function defect symptoms at the electric acupuncture group and model group,Longa score was no statistically significant difference(P>0.05)between the two groups;The EA group Longa score improved significantly after intervention,compared with model group,the difference was statistically significant(P < 0.05).2.Morris water maze experiment: From 4th to 8th day after modeling,the latency of water maze in model group and EA group is longer than sham operation group(P<0.05).Comparing with the model group,the latency of water maze decreased in electroacupuncture group(P<0.05).3.Western Blot results show: Beclin 1?LC3I/LC3 II protein expression of the rat hippocampus in electric acupuncture group is high than in the sham operation group(P<0.05);Compared with model group,the EA group of Beclin 1?LC3I/LC3 II in rat hippocampus protein expression was higher(P<0.05).4.RT-PCR results showed: comparing the three groups of rats hippocampus Beclin 1,LC3 II,Bcl-2 gene expression model group and the EA group of Beclin 1,LC3 IIgene expression were higher than in sham operation group(P < 0.05),but gene expression in the curative group was obviously higher in the model group(P < 0.05).Conclusion 1.Electric acupuncture at shenting and baihui can improve the nerve function and spatial learning and memory ability of MCAO rats;2.Electric acupuncture at shenting and baihui can regulate the gene and protein expression ralated to the autophagy network system of hippocampus tissue;3.Electric acupuncture at shenting and baihui is probably improve cognitive function in model rats by regulating the autophagy network system of hippocampus tissue.
