| Objective:Using the gene chip technology to the subjects fasting peripheral blood of transcriptomics studies,screening HIV/AIDS endoretention of damp heat syndrome of differentially expressed genes(m RNA),differential m RNA of gene ontology functional is classificed,participates in the inflammatory response of specific m RNA,using quantitative PCR techniques,verification of HIV/AIDS damp syndrome inflammation specific m RNA,inflammation of pathogenesis.Involvement in HIV / AIDS endoretention of damp heat syndrome inflammation gene molecular network building and TCM Syndromes of HIV/AIDS objective and efficacy evaluation to provide basic research,improve the transcriptome studies technology platform,provide a feasible method for the study of TCM syndrome.Methods:1 Experiment sample from a certain area of Henan Province,the 2011 national Ministry of Health issued the diagnostic criteria of Western medicine "AIDS treatment guidelines",the tutor group before the project of National Natural Science Foundation of China "AIDS TCM syndrome distribution rules and standards of syndromes of establishment and verification of"(Code: 90409004)research results "HIV / AIDS hot and humid intrinsic syndrome table for damp heat in HIV/AIDS intrinsic syndrome diagnostic criteria.2 Using Agilent single marker expression profile chip,using the Agilent feature extraction(FE)software version 9.5.3 data extraction,"TXT" file format,using SAS online analysis system,application independent sample t test for m RNA differential screening,to have gene expression.3 Using David(http://david.abcc.ncifcrf.gov/)online analysis of m RNA differentially to classific function annotation;using the relation between the string(http://string.embl.de)on the function of the different m RNA were analyzed,using the KEGG pathway database on m RNA differential pathway analysis.4 using quantitative polymerase chain reaction(PCR)analysis and validation in HIV/AIDS endoretention of damp heat syndrome inflammatory reaction of specific m RNA.Results :1 40 cases were included in the study,which HIV/AIDS hot and humid intrinsic syndrome in 24 cases,the same area of healthy people in 16 cases,the two groups in age,gender composition have not significant difference;CD4+ dampness heat syndrome group is lower than that of the healthy control group.The results showed that significant differences;CD8+ dampness heat syndrome group was higher than that in healthy control group,the result is significant difference.2 The use of spss21.0 software,t test analysis of m RNA between the two groups,and combined with the change of times numerical FC≥2 or FC≤0.5,177 m RNA were got the difference of the two,according the DAVID to functional annotation by three aspects:involved the biological processes(Biological Process,BP),Molecular Function(molecular function MF),cellular components(Cellular Component CC),which involved in the biological process of 77 kinds of categories,including participation in the inflammatory response(inflammation)have 9:CCL3↓,UNC13D↓,CR2↓,DARC↓,CCL3L3↓,IL6R↑,C2↓,PTAFR↑,AHSG↓,and inflammatory response involved to wounding,defense response,immune response,leukocyte mediated immunity,chemotaxi and other related functions.3 Using STRING(http://string.embl.de)to get the 35 sides of the composition of the genetic relationship,through the KEGG_PATHWAY pathway analysis,draw the passage 19:Staphylococcus aureus infection,hematopoietic cell lineage,Cytokine-cytokine receptor interaction,toll like receptor signaling pathway,Salmonella infection,chemokine signaling pathway,rheumatoid arthritis,complement and coagulation cascades.4 Using RT-PCR to validate the obtained CT value,in the hot and humid intrinsic group and the healthy control group CT values of relative quantitative comparison,IL6 R,CCL3 and C2 gene expression was up-regulated,which IL6 R trend with the chip results,CCL3 and C2 gene variation trend with the chip results are not consistent.Conclusion:1 Gene chip technology can get HIV/AIDS endoretention of damp heat syndrome and healthy control group,the m RNA differential expression profile to conduct the research to the biological basis of TCM syndrome.2 Differential gene for functional classification of ontology,come to participate in inflammatory response genes of nine,mainly involved to inflammation,trauma response,defense response,immune response,chemotaxis and other related functions,with hot and humid intrinsic was closely related with the inflammatory response mechanisms of card.3 Differential m RNA Involving on signaling chemokine signaling pathway,toll like receptor signaling pathway,complement and coagulation cascades.theory discussion on these pathways may participate in damp heat syndrome of inflammatory response.4 The complement and coagulation cascades level and HIV/AIDS hot and humid intrinsic syndrome are closely related,the complement system activation can activate mast cells,histamine release,caused vasodilation and increased permeability,these common clinical symptoms may with the AIDS of damp heat intrinsic inflammatory response mechanisms of card are close related. |
