Experimental Study On The Effect Of Tongluo Zhuanggu Pill On Lipid And Bone Gla Protein Calcitonin With Steroid-induced Avascular Necrosis Of Femoral Head In Rabbits

Objective:The model of Steriod-inducd avascular necrosis of femoral head of rabbits was induced by the horse serum and hormone,to observe the effect of Tongluo Zhuanggu Pill on the lipid and bone gla protein calcitonin indexes of rabbits of hormonal avascular necrosis of femoral head.Probing into its working mechanisms of preventing and curing of hormonal avascular necrosis of femoral head,and discuss its working mechanism and feasibilty of curing of hormonal avascular necrosis of femoral head,and provide theoretical basis for clinical application of Tongluo Zhuanggu pill.Methods:The experiment used 26 healthy adult New Zealand rabbits as the experimental object,choosing 9 rabbits randomly as the blank group,the others were the model group.Ensure that the body weight and sex differences of two groups showed no statistical significance by test.The horse serum was injected 10ml/kg through ear vein to the rabbits of the model group after weighing,and the horse serum was injected 6ml/kg again after 3 weeks.The Methylprednisolone was injected 40mg/kg through enterooelia to the rabbits of the model group one times a day,for 3 days.The blank group was injected the isodose normal saline according to the weight.The penicillin was injected 100000 U one time of all rabbits by muscle to prevent infection one times a day,for 7 days.The model group and the blank group were selected 1 rabbits randomly sacrificed after 4 weeks to confirm the modeling successfully.And then the rabbits of the model group were divided into the treatment group and the model group randomly,each group of 8.Making the Tongluo Zhuanggu Pill dissolved in distilled saline 10 ml boiling into suspension liquid,to lavage the treatment group,s rabbits according to the dose of 0.3g/kg of one times a day.The other groups were injected the isodose normal saline.Weighing regular in order to adjust the dosage.After 8 weeks,all rabbits were sacrificed and taken femoral head to abserve their hape and separating the serum to detecte lipid and bone gla protein calcitonin.All the date were analyzed by statistical software SPSS18.0.Results:(1)The general information:The rabbits weight,hair,appetite,sensitive and mental status of the model group were not as good as the blank group.The above symptoms gradually were improved after the rabbits of the treatment group were cured 2 weeks,and the performance was close to the blank group at the sampling time.The symptoms of the model group were not improvement.(2)Under optical microscope: Osteoblasts were reduced,the trabecular bones becomed thinner and sparse,and the structure of it were disorder,fat cells and the number of the empty lacuna were increase of the model group.The blank group showed normal to confirm the modeling successfully.The number of the fat cells in the marrow cavity were normal,and the trabecular bones arranged rules and its structres was integrity of the model group.(3)The results of lipid and bone gla protein calcitonin: In the aspect of triglyceride and total cholesterol,the model group was higher the blank group significantly(P < 0.05),the treatment group was lower than the model group significantly(P<0.05),the treatment group compared with the blank group was no difference(P>0.05).In the aspect of bone gla protein calcitonin,the model group was lower then the blank group significantly(P<0.05),the treatment group was higher than the model group significantly(P<0.05),the treatment group compared with the blank group was no difference(P>0.05).Conclusion:The large dose of hormones can cause lipid metabolism disorders,the osteoblasts were in degeneration and necrosis,leading to femoral head necrosis.Tongluo Zhuanggu Pill can improve the capillary permeability,improve the local microcirculation of the femoral head,reduce the pressure of the bone and promote the dead bone repaired and the new bone regeneration by adjusting the content of lipid and bone gla protein calcitonin.The Tongluo Zhuanggu Pill has a significant role in treatment for the Steriod-inducd avascular necrosis of femoral head.
