Objective: Establish animal model of the massive defect of bone, and transplant the free bone near the ipsilateral medial vastus and rectus femoris muscle, try to searching the possibility of the massive free bone revascularization in heterotopia, provide the experimental basis for using free bone after ectopic transplantation to repaire segmental bone defect.Method:Choose 50 adult rabbits as the research object, takeself-controlmethod. Experimental group: cut out 1.5cm long femorals on the right, clean the surrounding soft tissue, the contents of the medullary cavity and periosteum and swashed by normal saline. Control group: cut out 1.5cm long femorals on left of the same rabbit, and boil it with high temperature. Transplant the massive free femoral near the ipsilateral medial vastus and rectus femoris, fixed the stump of femoral with steel plate. The rabbits were sacrifieed and the specimens were procured at 2, 4, 6, 8, 10 weeks after surgery for observation, chinese ink perfusion and CD34 immunohistoehemistry. Mierovessel density(MVD) were assessed in order to evaluate angiogenesis.Results:In the experimental group: Ink dye blood vessels and CD34 positive blood vessels are showen at 4 weeks postoperative. 4, 6, 8 weeks postoperatively, the amount of ink dye blood vessels and CD34 positive blood vessels are grow incrementally(P<0.05), and reach a peak in postoperative 8 weeks. At this point, the connective tissue membranes have wrapped the graft and hard to peel off. 4, 6, 8 weeks ostoperatively, The amount of ink dye blood vessels and CD34 positive blood vessels are more than in the control group(P<0.05).In the control group: Ink dye blood vessels and CD34 positive blood vessels are showen at 6 weeks postoperatively. 6, 8, 10 weeks postoperatively, The amount of ink dye blood vessels and CD34 positive blood vessels are grow incrementally(P<0.05).Conelusions: The experiment prove that massive free bone can complete revascularization by ectopic transplantation. Fresh free bone complete revascularization at the postoperative 8 weeks and is faster than boiling bone, and is more conductive to repair the massive defect of bone by replantation.
