Part one:Ectopic bone formation of the hydroxyapatite nanotube implanted into rat skeletal muscleObjective To investigate the ectopic bone formation ability of hydroxyapatite nanotubes after implanted into femoral muscle pouch in rats.Methods The hydroxyapatite nanotubes were implanted into the left femoral muscle pouch of ten rats. Bilateral operation was performed for self-control and the right femoral muscle pouch of ten rats were just open but implant nothing. After surgery, X-rays and CT were taken immediately after surgery,4-week,8-week,12-week postoperatively. The rats were sacrificed twelve weeks after surgery and the specimens were harvested to analyzed by pathology.Results Each rats had no sign for infection postoperation; In each observation period, the hydroxyapatite nanotubes could be detected but the very high density of bone could not be seen; the bone cartilage cells, capillary formation and degradation of hydroxyapatite nanotubes powder could not be found in pathology around the implant.Conclusion The ectopic formation ability of pute hydroxyapatite nanotubes is limited and need be conformed in further study.Part two:A model of acute osteomyelitis established by radius in rabbitObjective To inoculate different doses of staphylococcus aureus into rabbit radius, and investigate the appropriate dose to establish acute osteomyelitis, according to radiological and microbiological change.Methods 35 male New Zealand rabbits were recruited for bilateral radius surgery. Different doses of staphylococcus aureus ATCC25923(1×104CFU for group A,1× 105CFU for group B,1×106CFU for group C,1×107CFU for group D,1×108CFU for group E, and 1×109 CFU for group F) and the 0.1ml saline were inoculated as experimental group A-F and control group. After surgery, X-ray and CT were taken immediately after surgery,2-week, and 4-week postoperatively. The radiographic score, the visual inspective score and the histological score were estimated four weeks postoperatively. The rabbits were sacrificed four weeks after surgery and the specimens were harvested for pathologic analysis and bacterial cultivation.Results Anorexia and lassitude were observed in all rabbits. Two rabbits died in group F died from infection and thus more excluded from analysis. Body weight loss was presented in group D,E and F. Radiographic, visual inspective and histological evidence of osteomyelitis were seriously appeared in group E and F, but mildly observed in group D. The radiographic score, the visual inspective score and the histological score of group E and F, was statistically higher compared with other groups. Bacteria was cultivated in group D,E and F, but not in other groups after bacterial culture.Conclusion The suitable dose of staphylococcus aureus ATCC25923 should be 1× 108CFU. The method of this investigation is convenient and satisfactory to establish the radius osteomyelitis model in rabbits.
