Objective: To explore the changes of peripheral blood T lymphocyte subsets and its significance in the terminal ileitis.Methods:90 SD rats were randomly divided into three groups, group M: model group, group S:suture group, group C: control group.Three groups have taken a different processing. Group M: the distal ileum and proximal colon were to be inosculated, manufacturing colon and ileal reverse flow model; Group S: experienced same degree of intestinal injury as the group M in the ileum and colon of the same position. Group C: only anesthesia was administered, without performing surgery. Three groups of rats were fed and cared for the same. 2, 4 and 8 weeks after surgery, 10 rats were randomly selected from each group to draw blood from the vein and ileum tissue samples were removed.Venous blood was to detect T lymphocyte subsets by FCM( flow cytometry), the ileum tissue was made into paraffin sections and scored.Results:1. The changes of the peripheral blood T lymphocyte subsets:(1) DNT cells: the proportions of DNT cells were significantly increased in group M and group S, group M were more apparent(P<0.05), having reached its climax in the 2nd week, indicating that DNT cells may be involved in the acute inflammatory reaction process.From the 2nd week to 8th week, numbers gradually declined, group S almost returned to normal level at 4th week, group M declined slower than the group S, there was significant difference between the two groups(P<0.05). It indicated that the DNT cells changed most obviously in the acute phase(0-4 weeks) of experimental terminal ileitis.(2) T4 cells: the proportions of T4 cells were significantly decreased in group M and group S, group M were more apparent(P<0.05), having reached its climax in the 2nd week. From 2nd week, group S has began to increase and reached normal levels at the 4th week.Group M remained at low levels.There were a few numbers even lower than the 2nd week.Group M recovered to nomal levels after the 4th week.It indicated that T4 cells may play an important role in the acute phase(0-4 weeks) of experimental terminal ileitis.(3) T8 cells:to the 2nd week,the proportions of T8 cells were significantly increased in both group M and group S, group M were more apparent(P<0.05).Up to the 4nd week, group M increased further,but the group S reached normal level, there was significant difference between the two groups(P<0.05). Group M returned to normal level at 8th week. It indicated that T8 cells may play an important role in the 4th to 8th week of experimental terminal ileitis.(4) DPT cells: the proportions of DPT cells were significantly increased in group M,having reached its climax in the 4th week.It has gradually declined at 4th to 8th week, still higher than group C at 8th week, group S had no significant changes during the entire time. there was significant difference between group M to group S(P < 0.05). It indicated that T8 cells may involve in the whole process of terminal ileitis(0 to 8th week).2. The correlation of peripheral blood T lymphocyte subsets and the ileum tissue inflammation degree:According to the observation of the obvious damaged ileum tissue,we found that,with extended time, the histopathologic grading was higher and inflammation reaction more serious. Through the correlation analysis of peripheral blood T lymphocyte subsets and the ileum tissue inflammation degree, it is certified that there was a positive correlation between the proportions of DNT,T8 and DPT three types of cells to related to ileum tissue inflammation severity(R value is 0.518?0.482?0.714,P < 0.05), there was a negative correlation between the proportions of T4 cells to the ileum tissue inflammation severity(R=-0.506,P < 0.05).Conclusions:1. In the experimental terminal ileitis,the proportions of DNT,T8 and DPT three types of T lymphocyte subsets increase, the proportions of T4 lymphocy decrease.All peripheral blood T lymphocyte subsets may be involved in the ETI.2. There is a positive correlation between the proportions of DNT,T8 and DPT three types of cells related to the ileum tissue inflammation severity(P < 0.05), there is a negative correlation between the proportions of T4 cells and the ileum tissue inflammation severity. T lymphocyte subsets may be as an important index of monitoring terminal ileum inflammation degree.
