Objectives: The research aimed to discover stromas proteins in gastric mucosal atypical hyperplasia and to understand possible mechanisms of microenvironment effect occurrence of early stage gastric cancer.Methods: 1. The stromas of normal gastric mucosa tissue(NGM) and gastric mucosal atypical hyperplasia(GMAH) was respectively purified from fresh NGM and GMAH tissues by laser capture microdissection(LCM), and obtained high purity stromas of NGM and GMAH. Then, the differentially expression proteins between NGM and GMAH tissues were identified by i TRAQ-tagging combined with 2D LC-MS/MS. 2. The cross-talk between stromas proteins in gastric mucosal atypical hyperplasia was analyzed by bioinformatics. 3. The expression of S100A6 and SOD3 in NGM and GMAH tissues was detected by Western-blot and immunohistochemistry staining.Results: 1. 165 differentially expressed proteins were identified from stromas of NGM and GMAH tissues, 99 proteins were up-regulation, 66 down-regulated in GMAH tissue. 2. These proteins were involved in cancer-associated signaling pathways such as p53, MAPK and cell cycle and apoptosis signaling pathways. These proteins in gastric mucosal atypical hyperplasia were associated with cellular growth, cellular proliferation, metabolism, apoptosis, translation, metabolism, protein transport, localization and humoral immune response. They were the nodes in the STRING network and between mutual relations. 3. The results of Western-blot and immunohistochemical staining showed that the SOD3 protein was low-expression and the S100A6 protein was over-expression in GMAH tissue(P<0.01).Conclusions: The research identified 165 differentially expressed stromas proteins in gastric mucosal atypical hyperplasia by laser capture microdissection combined with quantitative proteomics, of which 99 proteins were up-regulated and 66 proteins were down-regulated.
