| Objective: To study the expression level in different phases in gastric cancer,analyze the relationships between the two and provide theoretical basis for the research of the pathogenesis of gastric cancer and experimental basis,The expression levels of P53 and 8-OHdG in gastric cancer tissues,precancerous lesion tissues and chronic gastritis are detected.Method: 75 cases patients with gastric cancer that match groups(TNM staging of gastric cancer by UICC/AJCC standard(2010)),68 cases patients with precancerous lesion(26 cases of intestinal metaplasia and 42 cases of atypical hyperplasia),and 50 cases chronic gastritis were collected.Above all gastric cancer cases selected for the test period of newly diagnosed cases and wax block stored in the first hospital affiliated to hebei institute of north,using immune histochemical method(S-P)detection of P53 and 8-OHdG level in the diseased tissue and testing results for statistical processing and analysis of the relationship of P53 and 8-OHdG content in each group.At the same time all of patients were grouped by age,gender,the degree of differentiation,TNM stages and lymph node metastasis group and analyzed the relationship between the two patients and their clinicopathological features by statistical software SPSS22.0.Results:(1)P53 is mainly distributed in the nucleus,and the positive rate of patients with chronic gastritis,precancerous lesion and gastric cancer was gradually increasing.(P< 0.05);The expression level of P53 is not statistically significant in different age and gender of gastric cancer(P > 0.05).Patients with stage III or IV gastric cancer patients with P53 level obviously is higher than I,II,low level of P53,poorly differentiation of gastric cancer patients is significantly higher than high differentiation of gastric cancer patients,patients with lymph node metastasis with P53 level obviously higher than that of patients with no lymph node metastasis,the above differences are statistically significant(P< 0.05).(2)In precancerous lesions,P53 level in atypical hyperplasia was significantly higher than that of intestinal metaplasia,and the difference was statistically significant(P < 0.05).The positive rate of 8-OHdG in patients with chronic gastritis,precancerous lesion and gastric cancer was gradually increasing(P < 0.05).8-OHd G content was not statistically significant in different age and gender of gastric cancer(P > 0.05).Patients with stage III or IV gastric cancer 8-OHdG level is significantly higher than patients with stage I and II,poorly differentiated stomach cancer patients with 8-OHdG level obviously is higher than high school differentiation of gastric cancer patients,patients with lymph node metastasis with 8-OHdG level obviously higher than that of patients with no lymph node metastasis,the above differences are statistically significant(P < 0.05).(4)P53 expression level and 8-OHdG content were compared in gastric cancer tissues and precancerous lesion tissues,both of which were correlated.Conclusion:(1)8-OHdG,P53 level in acute or chronic inflammation of the organization,the stomach,the stomach before pathological tissues in positive level was gradually increasing trend,and in patients with stage III or IV gastric cancer tissue,poorly differentiated stomach cancer tissue,significantly higher in patients with lymph node metastasis with organization content,prompt both may participate in the development of gastric cancer,and malignant biological behavior and gastric cancer have a certain correlation;(2)8-OHdG and P53 expression were positively correlated,suggests there may be some kind of interaction relations,both common role in the occurrence of gastric cancer,evolve,monitoring,cancer of the stomach and the stomach before the lesion can be an objective index to early diagnosis,prognosis,specifically for both could effectively inhibit the development of tumor,improve the prognosis. |
