| [Objectives] The main objective of this paper is to detect the expression of Cathepsin D (CTSD), Heat Shock Protein 90a (HSP90α)and Epidemal growth factor receptor(EGFR) in Normal gastric mucosa tissues, Atypical hyperplasia tissue adjacent to carcinoma. Gastric Cancer (GC) and the lymph node metastasis tissues,Analysis the association of it’s expression levels and GC clinical pathological features Preliminary discussion the function of CTSD, HSP90a, EGFR in the process of GC’S occurrence,development, invasion and metastasis,Provide a experiment basis for GC’S clinical diagnosis, gene therapy and prognosis.[Methods] (1) The expression of Cathepsin D, Heat Shock Protein 90α and Epidemal growth factor receptor in Normal gastric mucosa tissues、 Atypical hyperplasia tissue adjacent to carcinoma、Gastric Cancer and the lymph node metastasis tissues is measured by immunohistochemistry and Insitu hybridization (2) By the chi-square test analysis the relationship between the expression level of CTSD,HSP90α、EGFR and the different stomach tissue,Compare the IHC and ISH test results by Kappa test.Using Rank correlation test to analysis the relationship between the expression level of CTSD,HSP90α、EGFR and clinicopathological features of GC patients.[Results] (1) In 110 cases of Normal gastric mucosa tissues,83 cases of Atypical hyperplasia tissue adjacent to carcinoma,110 cases of GC tissues and 78 cases of the lymph node metastasis tissues,The positive rate of CTSD protein and its mRNA respectively is 0、18.07%、80.91%、89.94%;its mRNA positive expression rate respectively is 0,7.22%,65.45%and 76.92%,and its CTSD protein expression level increases with gastric mucosa lesions from benign to tumor progression;Comparing Normal gastric mucosa tissues and Atypical hyperplasia tissue adjacent to carcinoma,the expression of CTSD and its mRNA is significantly increased.the expression in the lymph node metastasis tissues is higher in GC tissues. Spearman correlation analysis showed that shows the expression of CTSD protein is negativly correlated with tumor differentiation (r=-0.351,P<0.05),the higher of CTSD expression level,tumor differentiation worse.positively with the depth of tumor invasion,lymph node metastasis and TNM staging number (r=0.263、r=0.530、 r=0.523, P<0.05),the higher of CTSD expression level higher,the deeper the tumor infiltration depth,the more number of lymph node metastasis,higher TNM staging,but no with the patient’s age,gender and tumor size(P>0.05).(2) In 110 cases of Normal gastric mucosa tissues,83 cases of Atypical hyperplasia tissue adjacent to carcinoma,110 cases of GC tissues and 78 cases of the lymph node metastasis tissues,The positive rate of HSP90a protein and its mRNA respectively is 0.9%、5.66%、75.45%、89.74%;its mRNA positive expression rate respectively is 0、14.46%、60.91%,、 67.95%;Comparing Normal gastric mucosa tissues and Atypical hyperplasia tissue adjacent to carcinoma,the expression of HSP90a and its mRNA is significantly increased.the expression in the lymph node metastasis tissues is higher in GC tissues. Spearman correlation analysis showed that shows the expression of HSP90a protein is negativly correlated with tumor differentiation (r=-0.353,P<0.05),the higher of HSP90a expression level,tumor differentiation worse.positively with the depth of tumor invasion,lymph node metastasis and TNM staging number (r=0.440、r=0.340、 r=0.592, P<0.05),the higher of HSP90a expression level higher,the deeper the tumor infiltration depth,the more number of lymph node metastasis,higher TNM staging,but no with the patient’s age,gender and tumor size(P>0.05).(3) In 110 cases of Normal gastric mucosa tissues,83 cases of Atypical hyperplasia tissue adjacent to carcinoma,110 cases of GC tissues and 78 cases of the lymph node metastasis tissues,The positive rate of EGFR protein and its mRNA respectively is 0、12.05%、69.09%、84.62%;its mRNA positive expression rate respectively is 0、12.05%、60.91%、75.64%,and its EGFR protein expression level increases with gastric mucosa lesions from benign to tumor progression;Comparing Normal gastric mucosa tissues and Atypical hyperplasia tissue adjacent to carcinoma,the expression of EGFR and its mRNA is significantly increased.the expression in the lymph node metastasis tissues is higher in GC tissues. Spearman correlation analysis showed that shows the expression of EGFR protein is negativly correlated with tumor differentiation (r=-0.265,P<0.05),the higher of EGFR expression level,tumor differentiation worse.positively with the depth of tumor invasion,lymph node metastasis and TNM staging number (r=0.472、r=0.681、 r=0.507, P<0.05),the higher of EGFR expression level higher,the deeper the tumor infiltration depth,the more number of lymph node metastasis,higher TNM staging,but no with the patient’s age,gender and tumor size(P>0.05).(4) Spearman correlation analysis showed that the expression of CTSD, HSP90a and EGFR in GC tissues pairwise correlation, The expression level of CTSD and HSP90a are positively correlated in GC tissues, (r=0.853,P<0.05);The expression level of HSP90α and EGFR are positively correlated in GC tissues(r=0.734,P<0.05);The expression level of CTSD and EGFR are positively correlated in GC tissues(r=0.639,P<0.05)。[Conclusions] CTSD, HSP90a and EGFR may be associated with the malignant biological behavior Including tumor occurrence、development、 invasion and metastasis of GC。... |
