This essay concluded 4 parts:literature review,craft of drying,fingerprint of Rubia Cordifolia L.and charred Radix Rubiae,determination of five quinones compounds in Ru biae Radix et Rhizoma(carbonized),Rubia Cordifolia and charred Radix Rubiae on rabbit platelet aggregation,hemostatic mechanism in vivo,absorption Kinetics of Mollugin and Purpurin in Rats.1 Literature reviewThe literatures on medicine and herbs from ancient time to modern time were collec ted and studied,and the historical changes of methods processing Rubia cordifolia.L.and the survey of processed indifferent provinces were summarized.From Nanbeichao to Qi ng Dynasties,different processing methods were recorded in the literatures.A review for Rubia cordifolia.L.for traditional Chinese pharmaceutical use in medical history was pre sented for well documented works.And we summarize a review about clinical experience,chemical compositions,pharmacological.2 Chemical differences through processingThe compounds of Rubia Cordifolia L.and charred Radix Rubiae were compered by UPLC-MS.According to relevant literature and first class mass spectrometry,by UPLC-MS we indentified 9 compounds.3 Studies on HPLC fingerprint of Rubia Cordifolia L.and charred Radix RubiaeThe standard chromatographic fingerprinting of Rubia Cordifolia L.and charred Radi x Rubiae were established by the employment of HPLC technology respectively.The ana lysis was carried out with:GL Sciences Intertsil ODS-SP column(250 mm x 4.6 mm,5?m);The mobile phase were:0.07%-trifluoroacetic acid(A),Wavelength was288 nm and temperature was 30?.The injection volume was 10?l.We analysis 12 Rubia Cordifolia and 11 charred Radix Rubiae.The common peaks had been marked by standard of moll ugin and 2-methy-1,3,6-trihydroxymethy 1-9,10-anthraquinone.Which was analysized with t he Estimating System of Simlarity of 2004A Version(the Country's Pharmacopeia Comm ittee)on the Chinese Medicine Fingerprint Chromatography.The fingerprint chromatograp hy of Rubia Cordifolia and 11 charred Radix Rubiae.was estabilished.We can control t he quality of the Rubia Cordifolia before and after processed4 Simultaneous Determination of five quinones compounds in Rubiae Radix et Rhizo ma(carbonized)by HPLCTo establish an HPLC method for simultaneous determination of Purpuroxanthine,Pu rpurin,2-methy-1,3,6-trihydroxymethyl-9,10-anthraquinone,Rubimaillin in Rubiae Radix et Rhizoma(carbonized).The established method was rapid and simple with good accuracy and reproducibility for the determination of Rubiae Radix et Rhizoma(carbonized),the me thod was suitable for the quality control of Rubiae Radix et Rhizoma(carbonized).5 The Effects of Rubia Cordifolia L.and charred Radix Rubiae on rabbit platelet a ggregationThe results show that water extraction groups of Rubia Cordifolia and charred Radix Rubiae can increase ADP in contrast with alcohol extraction groups of Rubia Cordifolia and charred Radix Rubiae.Alcohol extraction groups of Rubia Cordifolia and charred R adix Rubiae can inhibit ADP at high concentrations,but can promote at low concentratio ns.The experimental results demonstrate that alcohol extraction groups of Rubia Cordifoli a and charred Radix Rubiae play role in bidirectional regulation.6 Hemostatic mechanism in vivo6.1 Clotting mechanism of acute blood-stasis ratsTo evaluate the effect of hemopoietic system in acute blood-stasis rats before and af ter processed by clotting four,hemorheology.Add the drug into rat to determine APTT,PT,TT,FIB and plasma viscosity and to observe the effect of hemopoietic system in ac ute blood-stasis rats.The experimental results demonstrate charred Radix Rubiae better th an Rubia Cordifolia in bleeding,but not so as Rubia Cordifolia in promoting blood circu lation.6.2 The effect of Rubia Cordifolia which cause of pathological injury of Lungs and the tongue of acute blood-stasis rats before and after processedWith additional research of pathological section,to understand the situation of lung and tongue tissues improvement of acute blood-stasis model rats after gastric perfusion.6.3 The effect of SOD,MDA,NO and NOS in lung tissues of acute blood-stasis rat sDrug groups can reduce the lung injury of acute blood-stasis model rats by our pre vious experiments.To discuss if the reason of lung injury caused by Oxidation-antioxidan t system balance disorder.7 Studies on the Intestinal Absorption Kinetics of Mollugin and Purpurin in RatsTo investigate the absorption kinetics of mollugin and purpurin intestines of rats.Est ablishing the in situ single pass perfusion model in rats,and adjusting the volume of rec irculation fluid by phenol red.Different quality concentration(12.33,24.66,49.32?g/mL)mollugin and(8.455,16.91,33.82?g/mL)purpurin of the absorption rate constant in the bowel segment of Peff has risen and they are all more than 0.2×10-4 cm/s.In the same mollugin element concentrations and purpurin element in the bowel period Colon>Duoden um>Ileum>Jejunum,and Peff is significant difference(P<0.05).Mollugin element and pu rpurin element in rat intestinal absorption mechanisms are the high permeability drugs,th e intestine at all have absorbed,and in the colon may exist specific absorption part.
