Objective: To study the chemical constituents in the ethyle acetate extracted for the75% aqueous Et OH extraction of Rubia cordifolia L.To provide a reference for the quality contronl of the product Er Xieting.Study the microscopic identification of stems,leaves and petioles of Rubia cordifolia L..Estabilish a pethod for HPLC fingerprint determining and the content of total triterpenes and oleanolic acid.It provides the theory basis for quality evaluation.Methods: The chemical constituents were sparated and purified by variety of chromatographic techniques.The structures of isolates were identified by various spectroscopic methods(NHR,spectrum,mass spectrometry)and literature compavison and so on.Using microscopic identification technology to study the pharmacognostic character of leaf,petiole and stem;The content of total triterpenoids was determined by vanillin chromatomety.Using oleanolic acid as the control,5% vanillin-glacialacetic acid-perchloric acid was used for color development,the absorbance was measured at557 nm by UV-Visible Spectrophotometry.The content of oleanolic acid in Rubia cordifolia L.from different habitats was determined by HPLC,the mobile phase was acetonitrile-0.4% phosphorice acid water(85:15),at an ultraviolet detection wavelength of 210 nm,the flow rate was 0.6 m L / min and a column temperature was 40?.Using oleanolic acid as the reference peak and gradient elution with the mobile phase composed of acetonitrile-0.4% phosphoric acid.The wavelength of detection was210 nm the flow rate was 0.6 m L / min and a column temperature was 40 ?.To compared the HPLC fingerprints from different main producting areas and the similarity evaluation.Results: Chemical investigation of Rubia cordifolia L.led to the isolation of 22 compounds,including Acetyloleanolic acid(1),1,6-dihydroxy-5-methoxylanthraquinore(2),2-hydroxy-3-methyl-anthraquinone(3),?-Sitosterol(4),(-)-episyringaresinol(5),trans-?-hydroxycinnamic acid(6),dibutylterephalate(7),(-)-secoisolariciresinol(8),(-)-3,4-divanillyltetrahydrofuran(9),lignans(+)-demethoxypinoresinol(10),(+)-syringaresinol(11),scopoletin(12),(+)-Isolariciresinol(13),1-(5-hydroxy-4-methoxyphenyl)-2-(5-hydroxy-3-methoxyphenyl)-1-propanone(14),bruselignan(15),Dihydrodehydrodiconiferyl alcohol(16),4,5'-dimethoxy-lariciresinol(17),(+)-7R,8S-5-Methoxydihydrodehydroconiferyl alcohol(18),5,5'-dimethoxy-7-oxolariciresinol(19),Rubifolic acid(20),Oleanolic acid(21),Ursolic acid(22).Established the microstructure characters of Rubia cordifolia L..The different main producting areas samples had a total triterpenes content of 0.5260% ~ 1.052% and a oleanolic acid content of 0.0508 % ~ 0.1050 %.Establised the HPLC fingerprint and the frinegerprint consisted of 18 common peeks.Consults: 22 compounds were isolated from the Rubia cordifolia L.,including 1 new compounds(14).This is the first time to report all of the compounds and compound1-3?5-19 were first isolated from Rubia Linn.Results it is exclusive in microscopic characteristics of Rubia cordifolia L.,to provide the evidence of identification.The established methods are sutabe for the determination of total triterpenoids,oleanolic acid of Rubia cordifolia L.,and the methods has reliable and reproducible.
