The Protective Effect Of Liangxue Huayu Recipe (LHR) On Experimental Liver Injury And The Related Molecular Mechanism Against Apoptosis

This thesis is composed of two aspects which include theoretical and experimental research on the protective effect and the molecular mechanism of Liangxue Huayu Recipe(LHR)for experimental liver injury.Liangxue Huayu Recipe(LHR)is set up.for the treatment of severe hepatitis.Severe hepatitis belongs to the concept "acute jaundice" in traditional chinese medicine.Theoretical study mainly involve five aspects:the history of Liangxue Huayu creating Method,the basis and mechanism of Liangxue Huayu Recipe's protective effect on acute jaundice,the principles which are applied to treat acute jaundice,and the related synergy significence of experimental recipe design.Damp-heat epidemic toxin,toxic heat flourishing and blood stasis are the main pathological factors of acute jaundice.Therefore,the principles of Liangxue Huayu Recipe(LHR)on the prevention and treatment of acute jaundice are established.The efficacy of Liangxue Huayu Recipe(LHR)which could clear heat and cool blood,promote blood circulation to remove blood stasis,nourish yin and strengthen the body resistance to the pathogenic factors,is the main mechanism of protecting body against acute jaundice.However,the pathogenesis is complex,so we should pay more attention to early diagnosis,dialectical treatment and prescription with the syptoms.Many experiments have been done to study the protective effect and the molecular mechanism of Liangxue Huayu Recipe(LHR)on liver injury,in vivo and in vitro,respectively,using rat models of acute liver injury(ALI)induced by D-GALN and LPS,and LO2 cell indcued by TNF-a and GalN.The results showed that Liangxue Huayu Recipe(LHR)improved the liver lesion and reduced the apoptosis of liver cells.This suggests that Liangxue Huayu Recipe(LHR)has the protective effect on experimental liver injury.The mechanisms are not only associated with alterations in mitochondrial membrane permeability,but also more importantly,with regulating apoptotic mediators in the mitochondrial pathway,such as the Bcl-2 family,cytochrome c and caspase-3.These findings provide a mechanistic explanation and identify a number of targets for the hepatoprotective action of Liangxue Huayu Recipe(LHR)by mitochondria.
