Effects Of Wenyang Jiedu Huayu Recipe On Intestinal Epithelial Cells Beclin1 And LC3-? In Rats With Acute-on-chronic Liver Failure

Objective:By observing the effects of Wenyang Jieduhuayu Granules on peripheral blood liver function and intestinal epithelial cell autophagyrelated proteins Beclinl and LC3-? in chronic-onset acute liver failure rats,explore its anti-liver failure mechanism.Methods:Of 60 male SD rats,6 were randomly divided into the normal group.The remaining 54 rats were sensitized with bovine serum albumin to establish a rat model of immune hepatic fibrosis,and then acutely challenged by intraperitoneal injection of D-Gal and LPS solution.A rat model of liver failure was established,and the normal group was injected intraperitoneally with the same dose of normal saline.The traditional Chinese medicine group was given Wenyang Jieduhuayu Granules by gavage,and both the normal group and the model group were given the same dose of distilled water by gavage.The liver function,the pathological changes of colon tissue,and the expression of autophagyrelated proteins Beclinl and LC3-? in intestinal epithelial cells were detected at 1h,12h,24h after acute attack in each group.Results:1.Liver function:Compared with the normal group,the other groups have differences between groups at each time point(P<0.01).The ALT and TBIL of the model group were significantly increased(P<0.01);compared with the model group,the ALT and TBIL of the traditional Chinese medicine group It decreased at all time points(P<0.01).2.Endotoxin:Compared with the normal group,the endotoxin level at each time point in the model group increased(P<0.01);compared with the model group,the endotoxin level at each time point in the traditional Chinese medicine group decreased(P<0.05).3.Histopathology of the liver:HE staining of liver tissue showed extensive damage to the liver tissue structure in the model group,with bridging and flaky necrosis of liver cells,and a large number of inflammatory cell infiltration in the portal area.4.Colonic histopathology:The surface of the intestinal mucosa of rats in the normal group is regular,clear in structure and normal in shape.In the model group,the intestinal mucosal epithelium was bleeding,necrosis,shedding,and inflammatory cells infiltrated.The intestinal mucosal epithelium of rats in the Chinese medicine group was intact,slightly congested,and a small amount of inflammatory cells infiltrated.5.The expression of autophagy-related proteins Beclinl and LC3-? in colon tissue:Compared with the normal group,the expression of Beclinl and LC3-? in the model group was significantly increased at each time point(P<0.01);compared with the model group,the expression of Beclinl and LC3-? at each time point in the traditional Chinese medicine group The expression of LC3-? was significantly reduced(P<0.01).Conclusion:1.In chronic-onset acute liver failure rats,the expression levels of autophagy proteins Beclinl and LC3-? in colon tissue are higher,and autophagy is in an activated state;2.Wenyang Jieduhuayu Granules can reduce intestinal mucosal damage by inhibiting the expression of autophagy proteins Beclinl and LC3-? in intestinal epithelial cells,which may be one of its anti-liver failure mechanisms.
