Objective: Through experim ental observation Ch inese medicine Xinjining Pill on adrenaline induced ventricular premature beat rhythm in the rat model of,the levels of serum nitric oxide(no)and serum levels of endothelial effe cts endothelin(ET-1),and explore the mechanism of the drug prevention of ventricular extrasystole,provide scientific theoretical basis f or eliminating dampness and phlegm,promoting blood circulation and removing blood stasis chamber of traditional Chinese medicine in the treatment of premature beat.Methods: 72 male Sprague Dawley(SD)rats(250-300g),SPF,adaptive feeding after a week,take 8 rats as a healthy control group(herei nafter referred to as the healthy group),the rem aining 64 only adre naline hydrochloride(100?g/kg)for modeling screening,eliminate is not sensitive to the drug and itself has arrhythm ia in rats,a total of 60 rats model was made successfully.Will meet the test requirements of 40 rats according to the random number table method were divided into 5 groups: model control group(hereinafter referred to as m odel group),Xinjining pill high dose group(1.948g/kg),Xinjining pill m iddle dose group(0.974g/kg),Xinjining pill low dose group(0.487g/kg)and propranolol group(2mg/kg),each group 8 rats in each.The concentration of gastric irrigation of Chinese medicine groups of rats with middle dose as the standard,the content of cr ude drug was 0.818g/m l.Rats in each group were administered orally for 1 times in the morning,each 2ml.The healthy group and model group were given the sam e dose of normal saline for 10 days.The 10 day last administered 1 hour after the rats were anesthetized fixed on the board and connection BL-420 F biological function experim ental system,the first recording electrocardiogram is norm al,then in the tail vein injection of rapid adrenaline hydrochloride induced rat arrhythm ia model recorded arrhythm ia appeared tim e,arrhythmia sustained changes in time,the heart rate of th e rat,and statistics of each group rats ventricular premature beat incidence.Finally,from the celiac artery blood determination the content of NO and ET-1 in serum.Results:1.Influence of arrhythmia in rats: compared with the model group,Xinjining pill high?middle dose group and propranolol gr oup can significantly postpone the occurrence time of arrhythm ia,shorten the duration of ventricular arrhythm ia,significant difference has statistical significance(P<0.05).Xinjining pills high do se group and propr anolol group in arrhyth mia appeared time no significant d ifference(P>0.05),but the duration of the relationship of arrhythmia function is better than that of propranolol gr oup,significant difference(P < 0.05).2.On rat ventricular premature beat(VPB)rate effects: compared with the model group,Xinjining pill of high,m iddle and low dose group can reduce rat ventricular premature rate,with dose increase ventricu lar early occurrence rate is on a downward trend.3.The rats serum NO,ET-1 levels: com pared with the healthy control group,model group rats serum NO content was significantly reduced,ET-1 content increased significantly,there was significant difference(P<0.05).Compared with the m odel group,Xinjining pill of high,middle and low dose group and propranolol group can make rats serum NO content increased significantly,significantly reduced the content of ET-1,the differences of statistical significance(P < 0.05).Traditional Chinese medicine dose groups,Xinjining pill effect in the high dose group is superior to middle dose group(P<0.05),middle dose group the effect was better than that of low dose group(P<0.05),effect of rat serum NO and ET-1 contents also showed dose related trend.There was no significant difference betwee n the effects of Xinjining pill high dose group and propranolol group on serum NO and ET-1 content of rats(P>0.05).Conclusion:1.Xinjining pill can better agains t by arrhythmia in rats induced b y adrenaline,the arrhythmia appeared to d elay time,arrhythmia duration and significantly the incidence of low ventricular premature beat,and the size of the dose was positively correlated with,traditional Chinese medicine high dose group of antagonism best.2.Xinjining pill could obviously improve ventricular premature rats serum NO level,significantly reducing the level of serum ET-1,has a good role in the prevention of ventricular premature beat.The experimental results suggest that Xinjining Pill on adrenaline induced ventricular premature rat prevention and treatment mechanism,probably by promoting the synthesis of endogenous NO,reduc e the release of endogenous ET-1 and antioxidant mechanism to restore balance to achieve.
