Objective:The subject aims at probing into the expression and effect of MAL in COPD lung tissue.Method:This paper is divided into two parts. In the first part will screen medical records which have diagnosed lung cancer and treated by surgery in the Department of thoracic surgery of First Hospital Affiliated to Nanhua University between 2012-2015, no matter male and female. According to the 2011 COPD diagnostic standards of Chinese Medical bureau of respiratory diseases, combined with the medical records of patients, the subjects were divided into COPD group and normal lung function group, each group has 5 people, through the Department of Pathology, collected para-carcinoma tissue,all the lung tissue were taken far away from the focus at least more than 5cm, pathologically confirmed tumor invasion, record the patient's age,sex, smoking index main indicators and the lung function, using IHC to detect the expression and distribution of MAL in lung tissues. in the second part of the experiment, healthy male SD rats(weight:250 + 10 g) were randomly divided into normal control group(normal feeding for 4 weeks) and COPD model group(feed for 4 weeks, in the first day and the 14 th day were subjected to tracheal instillation and daily smoking cigarettes, for a total of 28 days)(n = 8), after 4 weeks,kill the rats and execute bronchoalveolar lavage, lavage fluid cell count the cells in lavage fluid, collect the lung tissues,observe the morphological changes in lung tissue specimens by routine pathological section and hematoxylin-eosin staining, and detect the expression and distribution of MAL by IHC,use ELISA to detect the expression level of MAL, TNF-?, IL-10 in lung tissue homogenate supernatant.Result:1.MAL was mainly expressed in small bronchial epithelial cells in human lung tissues. Compared with normal lung tissue, the expression of MAL in COPD lung tissue was significantly decreased, and the difference was significant(P < 0.05).2.the HE staining showed that complete structure lung tissue of rats in normal group, there is no infiltration of inflammatory cells,,such as lymphocyte etc,in small bronchi and bronchioles,but in COPD group,there is large infiltration of inflammatory cells in the lung tissue, alveolar, pulmonary mesenchyme and tracheobronchial, proliferation of airway smooth muscle, decrease of microvilli, significant damage to airway epithelial cells, small bronchial is particularly significant.3.BALF showed that the COPD model group was significantly higher than the control group, the difference was significant(P < 0.05), mainly in neutrophils and lymphocytes.4.IHC showed that the expression of MAL in the lung tissue of COPD group was significantly weaker than that in the normal group, and the difference was significant(P < 0.05).5.ELISA results showed that MAL decreased, IL-10 decreased, TNF- alpha content increased lung tissues homogenate supernatant of model group,which is compared with the normal control group, the difference was significant(P < 0.05).Conclusion:1.The expression of MAL decrease in both human and rat's COPD lung tissues.2.MAL participate in the rat's COPD pulmonary inflammatory response.
