Objective: To observe the clinical effect of Shaoyao Gouteng Muer Tang in preventing and treating chronic peripheral neurotoxicity caused by oxaliplatin in FOLFOX regimen chemotherapy,provide evidence-based medicine based on clinical expansion of Shaoyao Gouteng Muer Tang in preventing and treating chronic peripheral neurotoxicity caused by oxaliplatin through a randomized controlled clinical trial.Methods: 80 cases of patients with colorectal cancer diagnosed by pathology or cytology and needed to implement FOLFOX regimen chemotherapy were selected, including 33 cases of carcinoma of colon and 47 cases of rectal cancer, 56 patients after eradication, 13 patients after palliative fistula operation and 11 patients were not performed, 56 cases of postoperative adjuvant chemotherapy and 24 cases of late palliative chemotherapy, clinical stage: 38 cases in stage II, 31 cases in stage III and 11 cases in stage IV, the treatment group and the control group each 40 examples were randomized according to the number table method and the selected order. The treatment group received improved FOLFOX regimen chemotherapy with Shaoyao Gouteng Muer Tang,the control group with FOLFOX chemotherapy alone. Two groups of patients should avoided taking other drugs that may cause nerve damage, and instructed to avoid cold stimulation during the treatment. Made a record of circumstances chronic peripheral neurotoxicity, changes in hemoglobin,the TCM syndrome score, KPS score and grade according to the criteria of peripheral neuropathy every 2 cycles of chemotherapy,and analyzed the collected data by SPSS 21.0 statistical processing software.Results: The treatment group and the control group at the 1-2 level of chronic peripheral neurotoxicity incidence rates were 58.75% and 51.25%,and at the 3-4 level of chronic peripheral neurotoxicity incidence rates were 36.25% and 40% respecttively in the 2nd and4 th cycles. There was no significant difference incidence(P>0.05), the treatment group and the control group at the 1-2 level of chronic peripheral neurotoxicity incidence rates were 55% and 33.75%,and at the 3-4 level of chronic peripheral neurotoxicity incidence rates were 28.75% and 61.25% respectively in the 6th and 8th cycles. There were statistically significant differences between the two groups(P<0.05).The incidence of chronic peripheral neurotoxicity differed among the patients of the two groups after 8cycles of treatment,there were 12 cases in grade I of chronic peripheral neurotoxicity of the treatment group, 15 cases in grade II, 4 cases in grade III and 0 cases in grade IV,the incidence rate was 77.5%,while 7 cases in grade I, 7 cases in II, 18 cases in grade III and5 cases in grade IV of the control group,the incidence rate was 92.5%, the both methods were significantly different and the difference between the two groups had statistic significance(P < 0.05). Chronic peripheral neurotoxicity degree between the control group and the treatment group were also obviously different and the grade 1-2 chronic peripheral neurotoxicity incidence were 67.5% and 35%, grade 3-4 chronic peripheral neurotoxicity occurrence rate were 10% and 57.5%.The treatment group KPS score increased from 79.25 to 80.19,while the control group decreased from 80.50 to 77.47 after 8 cycles of treatment,P<0.05 according to statistical analysis. There was some statistics difference in the TCM symptom effect, P<0.01. In the treatment group, 1 cases were cured, 13 cases were markedly effective, 21 cases were effective,and 5 cases were ineffective, the total effective rate was 87.5,while in the control group, 3 cases were markedly effective, 15 cases were effective,and 22 cases were ineffective, the total effective rate was 87.5%.Conclusion: 1.Shaoyao Gouteng Muer Tang can effectively reduce the incidence rates of chronic peripheral neurotoxicity caused by oxaliplatin in FOLFOX regimen chemotherapy2.Chronic peripheral neurotoxicity caused by oxaliplatin in FOLFOX regimen chemotherapy is related to the accumulated dosage.3. Shaoyao Gouteng Muer Tang can effectively alleviate the symptoms of chronic peripheral neurotoxicity caused by oxaliplatin in FOLFOX regimen chemotherapy, improve the KPS score.4. Softening liver for calming endogenous wind,promoting blood circulation and freeing channel is one of the most important ways to treat chronic peripheral neurotoxicity caused by oxaliplatin in FOLFOX regimen chemotherapy in TCM.
