Objective To investigate the expressions of PDGF-C and PDGF-D in renal tissue of IgA nephropathy with different degree of interstitial fibrosis, to elucidate mechanism and significance of interstitial fibrosis.Methods According to Banff grade system 50 renal samples were divided into four groups control group and fibrosis groups (mild moderate severe). The expression of PDGF-C and PDGF-D in kidney were determined by immunohistochemistry. Collecting patients 24h urinary protein, blood urea nitrogen and creatinine at the same time before renal biopsy, and correlation analysis with pathological and immunohistochemical results.Results in the control group, PDGF-C mainly expressed in renal tubular epithelial cells(Mainly expressed in the medullary loop, distal tubule, collecting duct), Artery endothelial cells, expression weakly in renal tubular cells and interstitial. In the experimental groups, PDGF-C constitutively expressed in glomerular parietal epithelial cells, renal tubular epithelial cells, renal interstitial and arterial endothelial cells, in the control group, PDGF-D expression weakly in endothelial cells. In the experimental groups, PDGF-D mainly expressed in mesangial area, vascular endothelial cells, renal tubular epithelial cells and interstitial cells cytoplasm.The expressions of PDGF-C in IgA nephropathy nephridial was significantly difference than those in normal control group (P<0.05), the expressions of PDGF-C in mild group was increased than those in normal control group, the expressions of PDGF-C in moderate group was significantly increased than those in normal control group, the expressions of PDGF-C in serious group was significantly increased than those in normal control group, the expressions of PDGF-C in moderate group was significantly increased than those in mild group, the expressions of PDGF-C in serious group was significantly increased than those in moderate group, the expressions of PDGF-C positively correlated with interstitial fibrosis degree (r= 0.654, P<0.01)The expressions of PDGF-D in IgA nephropathy nephridial was significantly difference than those in normal control group (P<0.05), the expressions of PDGF-D in mild group was increased than those in normal control group, the expressions of PDGF-D in moderate group was significantly increased than those in normal control group, the expressions of PDGF-D in serious group was significantly increased than those in normal control group, the expressions of PDGF-D in moderate group was significantly increased than those in mild group, the expressions of PDGF-D in serious group was significantly increased than those in moderate group, the expressions of PDGF-D positively correlated with interstitial fibrosis degree (r= 0.571, P<0.01)Renal tissue PDGF-C and PDGF-D expressions positively correlated with blood urea nitrogen and serum creatinine(P<0.05), The expressions of PDGF-C and PDGF-D in IgA nephropathy nephridial tissue have no obviously dependability with 24 hour urine protein.Conclusion Our results suggest that increased tubulointerstitial expression of PDGF-C and PDGF-D may play a role in the development and progression of tubulointerstitial fibrosis in patients with IgA nephropathy.
