Purpose: Through scopolamine to establishe the mild cognitive impairment in pathological model,to observe the effects of Pilose Antler Peptide on memory learning ability,brain nerve factor and anti oxidative effect of the cognitive dysfunction model animal,to explore the mechanism of cognitive function improvement effect of Pilose Antler Polypeptide,and enrich the theory of traditional Chinese medicine in the prevention and treatment of cognitive dysfunction,thus can deepen the mechanism research on the prevention and treatment of cognitive impairment by traditional Chinese Medicine,and to provide a new way of thinking and basis for the development of related drugs.Method: Animals in each group were administered from the day of the experiment,The dosage of each animal in each group was:The positive group intragastric administration Piracetam Tablets 300mg/kg,PAP in low dose group 10mg/kg,middle dose group 20mg/kg,high dose group 40mg/kg,blank group and model group were given the same volume of pure water.On the eighth day,1 hours after intragastric administration.In addition to the blank group,other 5 groups of rats were given dexamethasone acetate(0.3mg/kg)to establishe the model of Yang Deficiency,blank group were given the same volume of pure water.The eighteenth day of the experiment,through the training of rats.The nineteenth day,according to2mg/kg intraperitoneal injection of scopolamine rats after administration of 1H,30 min later test.Outcome Measures:(1)Record the number of rats jumping error response;(2)Enzyme linked immunosorbent assay(ELISA)to detect the changes of brain nerve factor(BDNF,NGF)in animal brain tissue;(3)Enzyme linked immunosorbent assay(ELISA)to detect the changes of neural mediators and enzymes(5-HT,NE,ACh,AChE,ChAT)in the brain tissues of animals;(4)Enzyme linked immunosorbent assay(ELISA)to detect the changes of antioxidant indexes(SOD,MDA,GSH-Px)in animal brain tissues.Result:(1)The observation of cognitive disorder model rats jumping error reaction times showed that Pilose Antler Polypeptide can obviously reduce the rat step-down number of errors;(2)on related brain nerve growth factor detection showed that Pilose Antler Polypeptide can increase the expression level of BDNF and NGF in brain tissue of rat model of cognitive impairment;(3)to detect and learning and memory related neurotransmitters andmetabolic enzymes show that Pilose Antler Polypeptide can increase 5-HT,NE,ACh,ChAT,and reduce AChE expression level in brain tissue of rat models with cognitive deficits;(4)on Antioxidation index detection showed that Pilose Antler Polypeptide can increase the SOD and GSH-Px,reduce the MDA expression levelsin the brain tissue of rat model of cognitive impairment.Conclusion: Through experimental study show that Pilose Antler Polypeptide can significantly improve scopolamine induced mild cognitive disorder model rats learning and memory function,the mechanism may be related to the effect of Pilose Antler Polypeptide regulating related brain neural factors,neurotransmitters,metabolic enzymes and antioxidant factors.
