Background and Objectives: With the change of the diet and the increase of social pressure,Constipation may be one of the major problems of modern which seriously affects people’s health-related quality of life.The constipation crowd is easy to appear the disturbance of intestinal flora.Furthermore long-term constipation will induce a variety of diseases.Studies show that,in last five years,all kinds of constipation treatments appear many problems in the clinical research and application.Therefore,looking for a safe,mild and effective intervention is so important.A great number of literature shows that probiotics and dietary fiber can induce laxative effects on constipation significantly.So the purpose of the my study is to establish a standardized constipated model of SD rats which induced by loperamide and use probiotics,dietary fiber,probiotics joint dietary fiber to intervene the models to investigate the effect and mechanism of probiotics joint dietary.Methods: 1)5mg/kg loperamide was given to the SD rats for 5 days to replicate the model of constipation.The probiotics(2ml/a rat,i.g.),dietary fiber(1g/kg BW,i.g.),probiotics joint dietary fiber(2ml/a rat,i.g.)were used to intervene the models for 7 days.After 5 days,by observing the excrement character and detecting the fecal water content to determine whether the building of model was successful.Small intestinal transit was determined by the experiment of Carbon powder propulsion in the twelfth day.2)Using PCR-DGGE to detect the changes in intestinal flora diversity of each group;3)ELISA assay vasoactive intestinal peptide(VIP),substance P(P),Cl-,Ca+ of the rat colonic tissue.4)Western-blotting assay aquaporin 3(AQP3),C-kit of the rat colonic tissue.5)RT-PCR assay proteinkinase A(PKA),neurokinin(NK1)of the rat colonic tissue.6)Determine the activity of Na-K-ATPase by Na-K-ATPase activity test kit.Results:1)The constipated model was successfully established: In the fifth day,the stool of other groups showed the traits of granule and water content of feces showed a significant reduction(P<0.05)and normal group exceptly,.In the twelfth day,compared with the constipated model group,water content of feces and small intestinal transit rates increased significantly,however,the effect of probiotics joint dietary fiber group is more prominent(P<0.01).2)In the twelfth day,PCR-DGGE method was used to analyze the changes in intestinal flora diversity of each group.Compared with the normal group,the intestinal flora diversity of constipated model group was significantly reduced,and other groups were improved in different level.3)Compared with the constipated model group,the concentration of VIP、P、Cl-、Ca2+ in other groups was increased significantly and the effect of probiotics joint dietary fiber group was most obvious(P<0.05).The difference between probiotics group and dietary fiber group was not significant however there was significant difference between normal group and constipated model group(P<0.05).4)Compared with the constipated model group,the level of AQP3 and C-kit in other groups was improved significantly(P<0.05)and the effect of probiotics joint dietary fiber group was most obvious(P<0.05).5)The level of m RNA of PKA and NK1 was significantly recovery in each group other than constipated model group(P<0.05))and the effect of probiotics joint dietary fiber group was most obvious(P<0.05).6)Results compared with normal group,Na+-K+-ATPase activity depressed significantly in constipated model group(P<0.05).However,Compared with the constipated model group,Na+-K+-ATPase activity was higher level(P<0.05)and the effect of probiotics joint dietary fiber group was most obvious(P<0.05).Conclusions: 1)The constipated model was successfully established.2)Probiotics and dietary fiber can improve constipation in different level.In detail,they can significantly increase the concentration of VIP、P、Cl-、Ca2+,and improve the level of AQP3 and C-kit,also they can increase level of m RNA of PKA and NK1,and activated Na+-K+-ATPase.
