Background: Autophagy is a kind of "self devour" at the subcellular level cell,that is a highly conserved cellular degradation process.The cytoplasm and organelle are isolated through the bilayer membrane to autophagic vesicles.and then transported to the lysosome or vacuole is broken down into macromolecules for cell recycling.This is a kind of self-modifying cell behavior for the protection of the outside world to survive adverse conditions.From a global perspective,the incidence of thyroid cancer are continuing to rise,it has caused widespread concern in the medical community.Although anaplastic thyroid cancer accounts for only a small part of the thyroid cancer,but not in differentiated thyroid cancer-related deaths in the thyroid has accounted for a large part,not only the survival rate,but the prognosis is not ideal.Some types of anaplastic thyroid cancer treatment effect poor,single or in combination with conventional chemotherapy program has been unable to produce an effective therapeutic effect.In precision medical gradual rise today,treatment of anaplastic thyroid cancer is expected to be significantly improved.In the beginning of the formation of thyroid cancer autophagy can inhibit tumor growth and proliferation,and autophagy can improve refractory thyroid cancer for radiotherapy and chemotherapy sensitivity.However,among the tumor has formed,as the deterioration of the environment not conducive to the growth of tumors in tumor cell survival when autophagy also can play the role of promoting the survival of tumor cells.In addition to the different types of tumor cell autophagy vary,so autophagy for thyroid cancer in the end still play a protective role in promoting tumor cell death has yet to be further studied.Objective: The study took resveratrol sensitive and non-sensitive two anaplastic thyroid cancer cell lines THJ-21 T and THJ-11 T,by detecting them at different time points after being treated with resveratrol autophagy occurs or not,Research anaplastic thyroid carcinoma sensitivity to resveratrol relations with autophagy,which found that resveratrol autophagy process is played a protective effect on cancer cells or inhibition.Aims for the future anaplastic thyroid cancer treatment clinic provides effective treatment strategies,provide theoretical support to generate new methods,so as to anaplastic thyroid cancer treatment or even a cure to help.Methods: Cells were treated with resveratrol 0.5 hours,1 hour,1.5 hours,2 hours,4 hours,6 hours,8 hours,10 hours,12 hours,16 hours,24 hours,48 hours,and then the cells were collected,immunization fluorescence detection,detection indicators LC3 and Beclin1 double standard,according to test results from selected autophagy more active processing time point immunochemical detection cells,cells were detected in the expression of LC3 and Beclin1 choose the most active autophagy process time extracting protein western-blotting experiments,testing these two proteins LC3 and Beclin1 in expression in the cells after resveratrol,by comparison with the control group of normal cultured tumor cells,which can find the role of autophagy in anaplastic thyroid cancer susceptibility to resveratrol.Results: Immunofluorescence results showed that autophagy occurred in resveratrol tolerance THJ-11 T when treated by resveratrol,while resveratrol sensitive THJ-21 T cells after resveratrol treatment autophagy basically did not happen,suggesting that autophagy to cancer cells produce a protective role in helping cancer cells resistant to drugs,and according to the exact point in time analysis results can be found,THJ-11 T by 16 hours after drug treatment and 24 hours autophagy occurs more active.Results immunocytochemistry also prompted by resveratrol treatment,the expression of autophagy-related indicators THJ-11 T resveratrol increased tolerance of cell lines,and for resveratrol sensitive cell lines THJ-21 T autophagy related indicators almost no expression.Conclusions: Autophagy in anaplastic thyroid cancer drug resistance process,playing a role in protecting the cancer cells,and play a positive role in cancer drug resistance.Autophagy in cancer cells when the drug exposure did not start,in time by about 16 hours of drug treatment autophagy began to appear,and the continuing role of the drug for 24 hours when autophagy reached more active state.In THJ-21 T cell lines which extremely sensitive to resveratrol express less hardly observed autophagy and its related indicators,we can see that it is likely that because there is no autophagy,so THJ-21 T extremely sensitive to resveratrol.Thus,we found a refractory anaplastic thyroid carcinoma treatment potential ways.When chemotherapy is administered to the patient,even before the chemotherapy drugs used to the patient,the appropriate dose of autophagy inhibition drugs with medication are both used in patients,may play a therapeutic effect,thereby significantly improving the patient's prognosis,prolong survival time.
