Cervical cancer is the most common malignant tumor of female reproductive system,in recent years,the incidence of this disease is rising.Operation and postoperative radiotherapy and chemotherapy were performed in the early stage of clinical treatment,but the efficacy was limited.As an auxiliary means of chemotherapy,traditional Chinese medicine(TCM)has been widely used in clinical practice.Resveratrol,a plant antitoxin,is a polyphenolic compound with a wide range of pharmacological activities,such as anti-inflammatory,antioxidant,anti-tumor and anti-aging properties.The aging and degenerative organelles in eukaryotic cells are dependent on the intracellular autophagy lysosomal pathway for degradation,so as to prevent the aggregation of mutagenic substances and gene mutation.The role of autophagy in the inhibition of proliferation of He La cells in cervical cancer by resveratrol has been rarely reported.This study explored the induction of autophagy in He La cells by resveratrol in vitro.Objective:To investigate the inhibitory effect of resveratrol on the proliferation and autophagy of He La cells in cervical cancer in vitroMethods:Take the effects of different concentrations of Resveratrol on the He La cells of cervical cancer as the object of study.In vitro cultivation of cervical carcinoma He La cells,which were divided into control group(with the only added medium being RPMI-1640),experimental group(in which Res was added with concentrations of 5?mol/L,10 ?mol/L,20 ?mol/L,50 ?mol/L,100 ?mol/L).The concentrations of Res increased gradually and acted on the He La cells of cervical cancer separately.the rates of the inhibitory effect on the proliferation of each group of He La cells were determined by MTT method,the apoptosis of He La cells after being treated with different concentrations of Res was determined by Hoechst 33258,the expression of Atg5 in He La cells after being treated with different concentrations of Res was determined by immunofluorescence method,the expression of proteins p62 and Beclin-1 related autophagy was determined by Western blot method.Results:1.Compared with control group,the growth of He La cells which were treated by Res with different concentrations of5?mol/L,10?mol/L,20?mol/L,50 ?mol/L,100?mol/L in the experimental group were inhibited significantly,and the rates of inhibitory effect on the proliferation in He La cells increased gradually as the concentrations of Res increased(P<0.01);Decreased migration of cervical cancer cells,the apoptosis in He La cells increased significantly;2.Under the fluorescence microscope,more green fluorescence of Atg5 were distributed in cytoplasm of He La cells after being treated by Res with different concentrations of 30?mol/L and 100?mol/L;3.48 hours later Res treated He La cells,the expression of p62 decreased gradually with the increase of the concentration of Res,while the expression of Beclin-1 increased gradually.Conclusion1.The growth of He La cells in vitro which were treated by Res were inhibited significantly and the phenomena of apoptosis became more and more obvious.The effects was positively correlated with the dosage of Res.as the dose of Res increased,the effect of proliferation and apoptosis increased gradually,the migration of cervical cancer cells decreased.2.The autophagic activity is regulated by the expression of p62 and Beclin-1which were related to autophagy,Res increased the activity of autophagy by regulating the expression of p62 and Beclin-1which were related to autophagy,then inhibited the proliferation of He La cells and induced apoptosis,finally played a role of antitumor.
