Background and ObjectiveAcute leukemia is one of the most common malignant tumors in the blood system. It is usually expressed as the abnormal clonal proliferation of leukemia cells in bone marrow and other hematopoietic tissues,which is inhibited by the normal hematopoietic system.At present, chemotherapy or hematopoietic stem cell transplantation (HSCT)can effectively cure part of patients, but there are still a large portion of the patients are refractory or appeared relapse, which is associated with leukemic multi-drug resistance.And the lack of specificity of clinical medication, killing the leukemia cells will also cause damage to normal tissue, its side effects are very big. On the other hand,the cost of hematopoietic stem cell transplantation is relatively large, and the quality of life of patients with serious complications after transplantation is affected, after the allogeneic hematopoietic stem cell transplantation.Among them, severe graft versus host disease (GVHD) will endanger the patient’s life,and that restrict its clinical application to some extent. So it is an important task to search for new therapeutic drugs and therapeutic targets.Increased expression of c-MYC is observed in both Acute Myeloid Leukemia (AML) and T-cell Acute Lymphoblastic Leukemia (T-ALL).and high expression of C-myc protein was associated with the occurrence and prognosis of leukemia.Patients with high c-MYC expression had higher WBC counts and a higher percentage of CD34+ or CD33+ cells, as well as splenomegaly,liver infiltration, higher BM blasts, and lower CR rate.Activation or amplification of the MYC oncogene family is one of the most frequent events associated with cancer.c-MYC is frequently activated in acute myeloid leukemia(AML), and plays an important role in the induction of leukemogenesis.c-MYC is also frequently reported to be upregulated in acutelymphoblastic leukemia (ALL).This research was aimd to investigate the effects of resveratrol on the proliferation and apoptosis of leukemia, the understanding of the mechanism of its anti leukemia effect, The study was aimed to investigate the effect of resveratrol on proliferation and apoptosis of the Human T lymphatic leukemia cells (Hut78).To observe the effect of different concentrations of Res on the expression of c-myc protein in Hut78 cells, and to elucidate the mechanism of resveratrol against luekemia.Materials and methodsCulture of human T lymphoid leukemia cells (Hut78),the Human T lymphocytic leukemia cells was used in this study. CCK-8 assay was used to detect the cell proliferation that was treated by different doses of Res. Apoptosis was detected by flow cytometry that the Hut78 cells was treated by differentdoses and different times.Western blot was used to detect the changes of C-myc protein in Hut78 luekemia cells.ResultCCK-8 assay found that resveratrol significantly inhibited the proliferation of Hut78 cells in a certain range, and showed a dose time dependence, the Res effect on the Hut78 cells 24 hours after the half concentration inhibition rate that IC50 was 98.7 mol/L. Flow cytometry showed that resveratrol could induce apoptosis of Hut78 cells and increase with the increase of drug concentration. The apoptosis rate of the cell that was treated by 100 mol/L Res was (54.43±2.66)% in 24 hours, The apoptosis rate of the cell that was treated by 200 mol/L Res was (63.90±2.55)% in 24 hours. The apoptosis rate of the cell that was treated by 100 mol/L Res was (78.90±1.46)% in 48 hours.The apoptosis rate of the cell that was treated by200 mol/L Res was (91.97±+2.67)% in 48 hours.The expression of C-myc protein in cell was detected by blot Western method which was downregulated.ConclusionResveratrol has obviously antileukemia effect.The proliferation of acute lymphoblastic leukemia cells was significantly inhibited,and apoptosis was induced.The mechanism may be associated with the down-regulation expression of C-myc protein...
