Objective: To observe the effect of Resveratol on apoptosis and the expression of bal-2 and bax protein in articular chondrocyte in experimental OA of rabbit. To study the mechanism of Resveratol in the treatment of osteoarthritis.Methods: 36 New Zealand white rabbits were randomly collected six as group A (the control group). The model of OA of the knee was established with Hulth technique in the remaining 30 rabbits.8 weeks later, random selection of six rabbits took X-ray and pathology examination to prove the model successful. The remaining 24 rabbits were devided into 4 groups: group B (model control group),group C(Resveratrol intervention low dosage group),group D(Resveratrol intervention middle dosage group),group E(Resveratrol intervention high dosage group).Then group A,B received stomach irritation daily with Distilled water and group C,D,E received stomach irritation daily with Resveratrol solution (concentration is 60mg/ml) in different dosage for 6 weeks. Daily dosage of Group C,D,E were 30mg/( kg·d),60mg/( kg·d),120mg/( kg·d) respectively. Then the rabbits were sacrificed and lateral cartilage sections of right femoral medial condyle were analyzed by immunohistochemistry for bcl-2 and bax, TUNEL for apoptosis.Result: 1. The apoptosis rates of chondrocyte in group B,C,D,E were significantly higher than that of group A (31.64±4.05%,27.42±2.71%,22.92±3.57%,18.15±2.21%,5.62±1.72%, P<0.01) . The apoptosis rates of chondrocyte in group B,C,D were decreased compared with that of group B (P<0.05,P<0.01 and P<0.01 respectively) . Significant differences of apoptosis rate were found among group C,D,E (P<0.05) .2. The positive rate of bcl-2 expression in chondrocyte in group B,C,D,E were significantly higher than that of group A (P<0.01) . The positive rates of bcl-2 expression in chondrocyte in group B,C,D were higher compared with that of group B (P<0.01) . No significant difference was found in the positive rates of bcl-2 expression in chondrocyte among group C,D,E (P>0.05) .3. The positive rate of bax expression in chondrocyte in group B,C,D,E were significantly higher than that in group A (P<0.01) . The positive rates of bax expression in chondrocyte in group B,C,D were lower compared with that of group B (P<0.01) . No significant difference of positive rate of bax expression were found between C and D (P>0.05) , D and E (P>0.05) while significant differences of positive rate of bax expression were found between C and E (P<0.05) .4.The ratio of the positive rate of bcl-2 expression to that of bax ingroup A was higher than that in group B (P<0.01) . The ratio wasincreased in group C,D,E compared with group B (P<0.01) .Significant differences of the ratio were found among group C,D,E(P<0.05) .5. Pearson's correlation analysis showed that chondrocytes apoptosis rate was negatively correlated with bcl-2/bax ratio (P<0.01) and the correlation coefficient was—0.558.Conclusion: 1. The apoptosis ratio of chondrocyte in experimental OA of rabbits was higher than that in normal rabbits. Resveratrol can lower the apoptosis rate of chondrocyte in experimental OA of rabbit . Suppressing the excessive apoptosis of chondrocyte in experimental OA was one of the mechanisms of resveratol in the treatment of osteoarthritis.2. Resveratrol can suppress the excessive apoptosis of chondrocyte in experimental OA by means of which Resveratrol up-regulated the expression of bcl-2 and down-regulated the expression of bax, thus improved the ratio of bcl-2 to bax. This is one of mechanisms that Resveratrol regulated the apoptosis of chondrocyte in experimental OA.
