Object:o explore the immunosuppression effects on uterine tissue inflamm,studying the effect on CD206?Arg-1 of LPS inflammatory M2 macrophages by IL-4.Method:We randomly divide 120 Kunming mice into control(A)and experimental group,Experimental group were divided into group B(intraperitoneal injection LPS),group C(ingestion IL-4),group D(intraperitoneal injection LPS and ingestion IL-4);Each group was divided into 7?9 and 11 days,there are 10 mice of every time point.All mice in group A were injected with normal saline 0.2 ml on the 1st day and 3rd day;All mice in group B were injected with normal saline 0.2 ml on the 1st day and injected with IL-4 on the 3rd day;All mice in group C were injected with IL-4 0.2 ml on the 1st day and 3rd day;All mice in group D were injected with IL-4 0.2 ml on the 1st day and 3rd day.Among them,first days by intraperitoneal injection,third days using tail vein injection.The uterus of mice were extracted on the 7?9?11th day at 3pm.Then the experimental results were obversed,Detect the expression of CD206 in mice uterus tissue through immunohistochemistry.And use in situ hybridization to detect the m RNA expression of CD206 and Arg-1.ELISA was used to detect the content of Arg-1 in mice uteruses.Results:(1)Comparing with group A,the number of CD206+ macrophages and protein expression in the uterus of group B mice at different time points are remarkably decreased(P<0.01);comparing with group B,The number of CD206+ macrophages and protein expression in the uterus of group D mice at 7?9 days are remarkably increased after injection of IL-4(P<0.01),it is reach the maximum value at the 9?11days,Return to normal level.In various parts of the uterus,CD206+ macrophages are mostly concentrated in the endometrium,there have a trend that it migration to uterine muscular layer and the outer membrane over the time.(2)Comparing with group A,the expression and IOD of CD206 and Arg-1m RNA in the uterus of group B mice at different time points are remarkably decreased(P<0.01),comparing with group B,the expression and IOD of CD206 and Arg-1m RNA in the uterus of group D mice at 7?9days are remarkably decreased(P<0.01),Return to normal level at 9?11days.(3)Comparing with group A,The protein expression of Arg-1 in the uterus of group B mice at different time points is significantly decreased in B group(P<0.01).Comparing with group B,The protein expression of Arg-1 in the uterus of group B mice at 7?9days is significantly decreased in B group(P<0.01),Return to normal level at 9?11days.Conclusion:(1)IL-4 may induce alternative activation macrophages(AAM)produced in uteru tissue of mice;(2)IL-4 can induce the production M2 type macrophages to be counter the inflammatory reaction caused by LPS;(3)As time gose on,IL-4 can promote the migration of AAM in the uterus tissue of mice.
