Expression And Clinical Significance Of MiR-19a And DLC1 In Tissues Of Gastric Adenocarcinoma | | Posted on:2017-09-19 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Wang | Full Text:PDF | | GTID:2334330488959516 | Subject:Internal medicine (digestive) | | Abstract/Summary: | PDF Full Text Request | | Background and Objective Gastric carcinoma is one of the most common malignant tumors.The incidence of gastric carcinoma has occupied the fourth in the world,and ranked secondin the cancer related death.Along with the advance of medical technology and the change of people’s lifestyle and eating habits,it still shows a high incidence trend of gastric carcinoma in our country,and tends to be younger.Because of high incidence,difficulty of early diagnosis,poor prognosis of gastric carcinoma,looking for new tumor markers is imminent,which are not only used for early diagnosis of gastric carcinoma,but also might provide a reliable basis for treatment and evaluating.In recent years,mi RNAs becomes a research hotspot.A large of study had shown that micro RNAs(mi RNAs)were long 19-25 nucl-eotides small fragments of non coding RNA,and targeted m RNA 3’non rotating recording area(3’untranslated region,3’UTR) completely integrated or not.It involved in the regulation of cell proliferation,differentiation and apoptosis of basic activities by degrading the target m RNA or inhibiting itstranslation at a post transcriptional level.Some studies have showed that mi R-19 a have been high expression in ovarian cancer,lung cancer,colorectal cancer,bladder cancer and so on,and also closely related with cancer cell invasion,metastasis.Deleted In Liver Cancer 1(DLC1)was famous for the tumor suppressor genesis in a variety of tumors.Most experiments have shown that DLC1 involved in processes of infiltration and invasion of cancer cells through genetic hybrid or missing DNA methylation decreased expression.Bioinformatics analysis showedthat DLC1 was a target gene of mi R-19 a.But they both were rarely research in gastric carcinoma.Adenocarcinoma is the common pathological type of gastric carcinoma,so this experiment is mainly to research the gastric adenocarcinoma.This study useed real-time fluorescent quantitative PCR and Immunohistochemistry to analysis the difference expression of mi R-19 a and DLC1 in gastric adenocarcinoma and their relationship with clinical pathological parameters.we further analysised their correlation to clarify molecular mechanisms of the invasion and metastasis of gastric adenocarcinoma.These may provide a new molecular marker,new gene treatment and drug resistance research of gastric adenocarcinoma in clinical work.Materials and Methods We selected 61 cases of gastric adenocarcinoma tissues and normal tissues adjacent to the tumor which confirmed by pathology performed in the Department of the surgery operation of the First Affiliated Hospital of Zhengzhou University from April 2014 to September2014.All patients were not received preoperative chemotherapy and radiotherapy.we tested the expression level of mi R-19 a in gastric adenocarcinoma by q RT-PCR,and the expression of DLC1 was deteced by immunohistochemistry.we also analyzed the expression level and clinical pathological factors,and the relationship between them.Results1. mi R-19 a in gastric adenocarcinoma tissue expression level was significantly higher than that of normal tissues, and the difference is statistically significant(P < 0.05).The relative expression level of mi R-19 a was associated with lymph node status,degree ofdifferentiation and TNM stage.There was no statistical significance of the expression of mi R-19 a in different age,gender(P>0.05).And mi R-19 a under the ROC curve area is 0.951,sensitivity of87.3%,specificity of 92.3%.Thus it can be seen that mi R-19 a ingastric adenocarcinoma diagnosis have good accuracy.2. The higher expression of DLC1 in gastric adenocarcinoma(34.43%) was significantly lower than that in normal gastric tissues(70.49%),and the differences was statistically significant(P<0.05).There was no statistical significance of the expression of DLC1 in different age,gender(P>0.05).The positive expression in middle to poorlydifferentiated group was significantly lower than that in well differentiated group.The positive expression in group with lymph node metastasis was significantly lower than that in group without lymph node metastasis.The positive expression in stage III,IVgroup was significantly lower than that in stage I,II group.The differences was statisticallysignificant(P<0.05).3. There was a significant negative relation between the expression of mi R-19 a and prediction target genes DLC1 in gastric adenocarcinoma(r=-0.398, P<0.05)Conclusions1. mi R-19 a is high expression,and may play a role in cancer promoting gene in the development of gastric adenocarcinoma tissue.The expression of mi R-19 a in gastric adenocarcinoma is related to differentiation,lymph node metastasis and tumor stage,not with age,gender.2. mi R-19 a under the ROC curve area is 0.951.It is higher sensitivity and specificity suggests that mi R-19 a may have value in diagnosis of gastric adenocarcinoma.3. DLC1 is downregulated,and may play a suppressor role in the development of gastric adenocarcinoma tissues.The expression of DLC1 in gastric adenocarrcinoma is related to differentiation,lymph node metastasis and tumor stage,not with age,gender.4.
